不同产地宽叶荨麻对人类风湿性关节炎滑膜成纤维细胞增殖和凋亡的影响

Effects of Urtica laetevirens from different areas on cell proliferation and apoptosis in human rheumatoid arthritis synovial cells

目的研究不同产地宽叶荨麻对人类风湿性关节炎滑膜成纤维细胞MH7A增殖和凋亡的影响。方法分别以庄河、鲁甸、阿坝、巴株产宽叶荨麻70%乙醇提取物处理MH7A细胞,24 h后用MTT法检测细胞增殖率。通过测定DNA片段化检测细胞凋亡率。结果 4种产地的宽叶荨麻都显示了一定的抑制MH7A细胞增殖的作用。其中鲁甸产宽叶荨麻显示了较强的抑制作用,因此进一步考察了鲁甸产宽叶荨麻不同组分对MH7A细胞增殖的影响,其中经大孔吸附树脂吸附以80%EtOH-H2O洗脱部分(LD80)显示了较强的抑制MH7A细胞增殖的作用。DNA片段化分析结果表明,LD80能显著引起MH7A细胞凋亡。结论鲁甸产宽叶荨麻能够抑制MH7A细胞增殖和诱导MH7A细胞凋亡。

ObjectiveThe antiproliferative and apoptosis inducing effects of 70% EtOH extract from Urtica laetevirens from different areas on MH7A human rheumatoid arthritis synovial cells were test in this study. MethodsMH7A cells were treated with different concentrations of 70% EtOH extract of Urtica laetevirens from Zhuanghe,Ludian,Aba and Bazhu.The antiproliferative activity was tested against MH7A cells using the MTT assay,and the apoptosis was assessed with DNA fragmentation after 24 h. ResultsThe 70% EtOH extract of Urtica laetevirens from Zhuanghe,Ludian,Aba and Bazhu inhibited cell proliferation in MH7A cells.And the 70% EtOH extract of Urtica laetevirens from Ludian has a stronger effect on cell proliferation.Therefore we investigated the effects of different elution fractions from 70% EtOH extract of Urtica laetevirens from Ludian on MH7A cell proliferation.We found that 80% EtOH-H2O elution fraction（LD80） showed stronger antiproliferative activity against MH7A cells.LD80 could induce DNA fragmentation in MH7A cells. ConclusionWe found the extract of Urtica laetevirens in Ludian could inhibit cell proliferation and induce apoptosis in MH7A cell.
