基于抗炎镇痛作用的气滞胃痛颗粒有效组分活性研究 被引量:20

摘要 目的在气滞胃痛颗粒抗炎镇痛有效组分体内外筛选研究的基础上,展开该复方中7个主要组分与抗炎镇痛活性相关性分析研究,从化学组分群的层次上揭示气滞胃痛颗粒抗炎镇痛药效物质基础。方法采用L16(215)正交设计表将气滞胃痛颗粒中7个主要组分制成16个配方(剂量均按原组方生药量折算),观察各组小鼠的扭体次数、足肿胀度和耳肿胀度,以扭体抑制率、足肿胀抑制率和耳肿胀抑制率为抗炎镇痛活性评价指标,对活性成分与药理评价指标进行统计学分析。结果以扭体抑制率为评价指标,气滞胃痛颗粒镇痛有效组分活性大小为:白芍总苷>柴胡总皂苷>延胡索总生物碱(P<0.05);以足肿胀抑制率为评价指标,气滞胃痛颗粒抗炎有效组分活性大小为:延胡索总生物碱>甘草总黄酮>柴胡总皂苷(P<0.05);以耳肿胀抑制率为评价指标,气滞胃痛颗粒抗炎有效组分活性大小为:延胡索总生物碱>柴胡总皂苷>甘草总黄酮>白芍总苷(P<0.05)。结论气滞胃痛颗粒中有效组分的不同配方对疼痛和炎症模型表现出不同的作用,该研究确定了各组分与抗炎镇痛活性的相关程度,为气滞胃痛颗粒有效组分量效关系的深入研究奠定基础。 ObjectiveTo study the correlation between the 7 main effective components in Qizhiweitong Granules and their anti-inflammatory and analgesic activity on the basis of in vivo and vitro screening,revealing the substance basis of anti-inflammatory and analgesic effect of Qizhiweitong Granules at the level of chemical component group. MethodsAccording to the orthogonal design table L16(215),sixteen chemical component formulae were compounded from 7 components in Qizhiweitong Granules(the dose according to the original complex prescription).The times of body twisting,the swelling degree of foot and ears of the mice were observed.Writhing resistant rate,foot-swelling resistant rate and ear-swelling resistant rate were taken to evaluated anti-inflammatory and analgesic activity.Statistical methods were used to analyze the relation between active components and pharmacological evaluation index. ResultsUsing writhing resistant rate as evaluation index,the order of analgesic effective components in Qizhiweitong Granules was: glucosides of Paeony saponins of Radix Bupleuri alkaloids of Corydalis(P0.05).Using foot-swelling resistant rate as evaluation index,the order of anti-inflammatory effective components was: alkaloids of Corydalis flavonoids of Glycyrrhizaesaponins of Radix Bupleuri(P0.05).Using ear-swelling resistant rate as evaluation index,the order of anti-inflammatory effective components was: alkaloids of Corydalissaponins of Radix Bupleuri flavonoids of Glycyrrhizaeglucosides of Paeony(P0.05). ConclusionThe different compatibilities of chemical component groups in Qizhiweitong Granules have different effects on pain and inflammation mode1.This study defined the correlation between effective components and anti-inflammatory and analgesic effect and laid the foundation for dose-effect relationship of active components in Qizhiweitong Granules.
