摘要
目的:研究Eag1钾通道在前列腺癌(PCa)组织中表达的状况,为探讨其与PCa发生、发展机制的相关性及能否作为PCa的诊治靶点提供理论依据。方法:采用RT-PCR和免疫组化的方法,检测Eag1钾通道mR-NA和蛋白在雄激素依赖性前列腺癌(ADPCa)组织、雄激素非依赖性前列腺癌(AIPCa)组织和癌旁非瘤组织中的表达水平。结果:Eag1钾通道mRNA在ADPCa癌旁非瘤组织(A)、AIPCa癌旁非瘤组织(B)、ADPCa组织(C)和AIPCa组织(D)中表达相对系数分别为0.265±0.413、0.167±0.511、2.673±2.988和2.815±2.901,与相应非瘤组织相比,Eag1钾通道mRNA在前列腺癌组织中表达显著增高(P<0.05);Eag1钾通道蛋白在A、B、C和D组中阳性表达率分别为7.4%、6.7%、88.9%和86.7%,与相应非瘤组织相比,Eag1钾通道蛋白在癌组织中表达显著增高(P<0.05)。结论:Eag1钾通道可能参与了PCa的发生和进展的病理生理学过程,有望成为PCa诊治中有价值的靶点。
Objective: To investigate the expression of the Eagl K( + ) channel in the prostate cancer (PCa) tissue, its correlation with the development and progression of PCa, and whether it could be a target for the diagnosis and treatment of PCa. Methods: We used RT-PCR and immunohistochemistry to determine the mRNA and protein expressions of the Eagl K( + ) channel in the normal peritumoral tissue of androgen-dependent PCa (ADPCa) (group A) and androgen-independent PCa (AIPCa) (group B) as well as in the tumorous tissue of ADPCa ( group C) and AIPCa ( group D). Results : The relative coefficients of the mRNA expres- sion of the Eagl K( + ) channel were 0.265 ± 0.413, 0. 167 ± 0.511,2. 673± 2. 988 and 2.815 ± 2. 901 in groups A, B, C and D, respectively, increased significantly in the latter two groups ( P 〈 0.05 ). The positive rates of the protein expression of the Eagl K ( + ) channel were significantly higher in groups C (88.9%) and D (86.7%) than in A (7.4%) and B (6.7%) (P 〈0.05).Conclusion : The Eagl K( + ) channel might be involved in the pathophysiological processes of PCa, and is expected to be a valuable target for the diagnosis and treatment of PCa.
