水蛭微粉对急性血瘀大鼠血液流变性的影响 被引量:6

摘要 目的:研究水蛭微粉对急性血瘀大鼠全血黏度和血浆血管性血友病因子(vwF)、纤溶酶原激活剂抑制物-1(PAI-1)含量的影响。方法:100只大鼠随机分为水蛭水煎剂0.54 g·kg-1组、水蛭普通粉0.54 g·kg-1组、水蛭微粉0.09 g·kg-1组、水蛭微粉0.18 g·kg-1组、水蛭微粉0.27 g·kg-1组、水蛭微粉0.36 g·kg-1组、水蛭微粉0.45 g·kg-1组、水蛭微粉0.54 g·kg-1组、空白对照组和模型对照组,每组10只,连续给药10 d后,采用肾上腺素加冰浴的方法复制急性血瘀模型,观察水蛭微粉对模型大鼠全血黏度及血浆vwF、PAI-1含量的影响。结果:与空白对照组比较,模型对照组大鼠的全血低切黏度、中切黏度、高切黏度、血浆vwF和PAI-1含量均明显升高(P<0.01或P<0.05);与模型对照组比较,水蛭微粉各剂量组可降低全血黏度及血浆vwF、PAI-1含量(P<0.01或P<0.05),水蛭微粉活血化瘀作用的等效剂量约为水煎剂、普通粉剂量的1/2~2/3。结论:水蛭微粉能够改善急性血瘀大鼠血液流变性,具有显著的活血化瘀作用,其作用机制可能与降低大鼠血浆vwF、PAI-1含量有关。 Objective : To study the effect of Hirudo micropowder on the whole blood viscosity and the contents of plasma von Will- ebrand factor ( vwF ), plasminogen activator inhibitor - 1 ( PAl - 1 ) in rats with acute blood stasis. Methods: 100 rats were randomly divided into Hirudo decoction 0. 54 g · kg-1 group, Hirudo powder 0. 54 g · kg- 1 group, Hirudo micropowder 0. 09 g · kg- 1 group, Hinldo micropowder 0.18 g · kg - 1 group, Hirudo micropowder 0. 27 g · kg - 1 group, Hirudo micropowder 0. 36 g · kg-1 group, Hirudo micropowder 0.45 g · kg-1 group, Hirudo micropowder 0. 54 g · kg-1 group, blank control group anti model control group, with !0 rats in each group. Acute blood stasis models were replicated with method of adrenaline and ice bath lOd after continuous administration to observe the effect of Hirudo micropowder on the whole blood viscosity and the contents of plasma vwF, PAl- 1 in rats with acute btood stasis. Results: The whole blood low shear viscosity, medium shear viscosity, high shear viscosity and cantents of plasma vwF and PAI- 1 of model control group were significantly increased ( P 〈 0.01 or P 〈 0. 05 ) compared with those of blank control group; the whole blood shear viscosity and the contents of plasma vwF, PAI- 1 ( P 〈0. 01 or P 〈0.05 ) of different dose groups of Hirudo micropowder were decreased compared with those of the model group; the equivalent dose of Hirudo micropowder promoting blood circulation mad removing Blood Stasis is about 1/2 to 2/3 common powder and decoction of Hirudo micropowder. Conclusion: Hirudo micropowder can improve blood rheology of rats with acute blood stasis, and it possesses obvious effect of promoting blood curculation amt removing blood stasis, the mechanism might be related to the content decrease of plasma vwF, PAI - 1.
