摘要
目的:检测Omp31-BLS融合肽抗血清的质量。方法:利用间接酶联免疫吸附实验、免疫双扩散实验以及凝集实验检测Omp31-BLS融合肽制备的抗血清的质量。结果:Omp31-BLS融合肽抗血清可以较好地识别Omp31-BLS融合肽蛋白质,而且,当抗体被稀释4 000倍的时候,酶联免疫吸附实验测定的OD值为1.2,明显高于其他3个对照组。在免疫双扩散实验中,抗血清被稀释64倍的时候,仍然能够出现明显的免疫沉淀线。此外,凝集实验也反映出Omp31-BLS融合肽抗血清可以与Omp31-BLS融合肽抗原发生强烈的凝集作用。结论:Omp31-BLS融合肽抗血清可以较好地识别对应的蛋白质抗原,二者之间存在明显的抗原-抗体反应,为利用Omp31-BLS融合肽进行布鲁氏菌病的血清学诊断提供了理论基础和技术支持。
Objective: this research is to detect the quantity of Omp31 - BLS fusion peptide antiserum. Method: Using the indirect enzyme linked immunosorbent assay, immune double diffusion test and agglutination test, the antiserum which was prepared by Omp31 - BLS fusion peptide was detected. Result: Omp31 - BLS fusion peptide antiserum could better identify Omp31 - BI.S fusion peptide. Meanwhile, in ELISA assay the optical density value was I. 2,when the antibody was diluted 4000 times. And it was significantly higher than other three groups. In immune double diffusion experiment, obvious immune precipitin line still appeared when the antiserum was diluted 64 times. Besides, agglutination test also reflected that strong agglutination phenomenon existed between Omp31 - BLS fusion peptide antise- rum and Omp31 -BLS fusion peptide. Conclusion:Omp31 -BLS fusion peptide antiserum could better identify the corresponding protein antigen, and the obvious antigen -antibody reaction was existed between Omp31 -BLS fusion peptide antiserum and Omp31 -BLS fusion peptide. And assay results provided theoretical basis and technical support for the brucellosis serological diagnosis using Omp31 - BIAS fu- sion peptide.
