摘要 目的:探讨钙网蛋白(CALR)在对甲氨蝶呤(MTX)耐药的人绒毛膜癌中的作用。方法:采用Western blot印迹和细胞免疫荧光方法检测CALR蛋白在对MTX间断诱导耐药绒毛膜癌细胞株JeG-3/MTXA1与亲本细胞JeG-3中的表达差异,观察利用RNA干扰(RNAi)技术干扰CALR表达后耐药细胞耐药指数的变化。结果:Western blot验证结果,CALR在耐药细胞JeG-3/MTXA1中的表达明显高于亲本细胞JeG-3。细胞免疫荧光结果,在耐药细胞JeG-3/MTXA1中,CALR蛋白表达明显较亲本细胞JeG-3增多,且近核表达增强。经过RNAi后,耐药细胞JeG-3/MTXA1的耐药指数下调74.05%(33.94±0.68vs8.81±1.58),而对细胞增殖无明显影响。结论:CALR蛋白在耐药细胞中明显上调,干扰其表达后耐药细胞可明显降低其耐药指数。CALR可能参与对MTX耐药绒毛膜癌的发生。 Objective:To investigate the activity of calreticulin (CALR)in the chemo-resistance of chorio- carcinoma to methotrexate. Methods:The expression of the CALR in MTX-resistant human choriocarcinoma cell JeG-3 was validated by western blot and immunocytochemistry. Ribonucleic acid interference(RNAi) was used to inhibit the expression of CALR. The resistant index was also detected before and after RNAi. Re- sults:CALR was found to be significantly up-regulated in MTX-resistant cells both in western blot and ICC. Besides the high intensity of CALR was exhibited in chemo-resistance cells,the translocation of CALR was al- so found. The resistance index(RI) decreased by inhibition of the expression of CALR(33. 94 ~0. 68 vs 8. 81 ~ 1.58),and there was no influence on cell generation cycle. Conclusions:The expression of CALR was significantly up-regulated in chemo-resistant cells, and the resistant index decreased dramatically by CALR-RNAi. CALR might be involvede in the chemo-resistance of choriocarcinoma to MTX.
