自噬调节与肿瘤治疗相关的研究进展 被引量:17

Regulation of autophage and its role in cancer therapy

摘要 目的:总结自噬及其调节机制的文献,探讨自噬与肿瘤的关系和调节自噬在肿瘤治疗中的作用。方法:应用PubMed文献检索系统和中国生物医学文献服务系统(SinoMed),以"自噬、肿瘤形成、耐药和放疗抵抗"为关键词,检索2007-01-2012-03有关自噬的发生和调节及其在肿瘤治疗中作用的文献。纳入标准:1)自噬的过程和调节机制;2)自噬与肿瘤的关系;3)调节自噬与肿瘤治疗。根据纳入标准,符合分析的文献37篇。结果:抑癌基因和自噬相关基因的突变常导致自噬调节障碍,并且与肿瘤形成密切相关。应用自噬抑制剂和基因干扰技术调节自噬使细胞内自噬活动维持在合适的水平,对于肿瘤的预防和提高放、化疗敏感性具有积极的作用。结论:自噬可以作为肿瘤治疗的潜在干预靶点,调节自噬能为肿瘤治疗研究提供新的策略。 OBJECTIVE:To summarize publications on the study of autophage and autophage regulation,and explore the relationship between autophage and caner as well as the role of autophage regulation in cancer treatment. METHODS: Publications from January 2007 to March 2012 on the study of autophage process and regulation and its role in cancer therapy were searched by key words of autophage, tumorigenesis, drug resistance and radiation resistance from the PubMed and SinoMed index system. Inclusion criterion.. 1) The process and regulation mechanism of autophage. 2) The relationship between autophage and cancer. 3) Autophage regulation and cancer therapy. According to the criterion,37 publications were studied and cited in this article. RESULTS: Mutation of tumor-suppressor gene and autophage-associat- ed gene usually leads to disorder of autophage regulation,which is closely related with tumorigenesis. Regulating autoph- age with autophage inhibitor and gene interference technology to maintain cell autophage in a right level is very important in cancer prevention and sensitizing tumor to radiotherapy and chemotherapy. CONCLUSION: Autophage is a potential in- tervention target in cancer treatment,and regulating autophage provides a new stratage in research of cancer therapy.
