RhoE在诱导人脑胶质细胞瘤凋亡中的作用 被引量:2

摘要 目的:研究和探讨RhoE在诱导胶质瘤细胞凋亡中的作用,为胶质瘤分子靶向治疗寻找新的靶点。方法:采用western blotting检测RhoE在人脑胶质瘤细胞系中的表达水平;构建重组pcDNA3.1-RhoE真核表达载体,将质粒快速转染U87细胞,采用四唑盐法检测细胞增殖能力的变化;Hoechst染色法观察细胞核的形态改变;流式细胞术检测细胞凋亡的情况;western blotting分析细胞Bcl-2、Bax表达变化,以及capase-3和NF-kB的活化水平。结果:RhoE蛋白在T98、A172和U87细胞系中表达明显低于正常人脑胶质细胞(P<0.05);上调RhoE表达后,U87细胞增殖能力明显下降(P<0.05),并且发生了显著的细胞凋亡改变(P<0.05);western bolt检测显示裂解的caspase-3水平增加,Bcl-2表达水平下降,Bax的表达水平升高,另外,p65在细胞核中的表达降低。结论:RhoE可能通过抑制NF-kB的活性,下调Bcl-2/Bax的比例,从而促进caspase-3的活化,最后诱导U87细胞发生凋亡。
