摘要
目的:通过对衰老模型大鼠进行多重慢性应激,观察多重慢性应激对大鼠海马糖皮质激素受体mRNA(GR-mR-N A)表达的影响,同时,对多重慢性应激下衰老模型大鼠进行穴位埋线,探讨穴位埋线对衰老的干预作用。方法:健康3月龄SD雄性大鼠48只,随机分为4组:空白对照组、D-半乳糖组、复合衰老组、复合衰老加埋线组。空白对照组:仅给予6周腹腔注射生理盐水。D-半乳糖组:仅给予6周腹腔注射D-半乳糖。复合衰老组:给予6周腹腔注射D-半乳糖,同时从第3周开始给予制动应激,应激每天1次,共应激4周;从第3周开始进行噪声刺激,每天1次,每次30min,共4周;从第3周开始,进行昼夜颠倒的光刺激,共4周。复合衰老加埋线组:造模方法同复合衰老组,另外每周埋线1次,共6周。实验结束后断头,并取出约20mg海马组织,测量每只大鼠海马糖皮质激素受体mRNA表达。结果:各组大鼠海马糖皮质激素受体mRNA表达的变化:与空白对照组相比,D-半乳糖组、复合衰老组海马的GR-mRNA基因表达均减少(P<0.05)。复合衰老组与D-半乳糖组比较,差异有显著性意义(P<0.05);复合衰老加埋线组与复合衰老组比较,差异有显著性意义(P<0.05)。结论:复合衰老组的海马糖皮质激素水平过高,下调了GR-mRNA的表达,其受损较D-半乳糖组更为严重,穴位埋线可以调节复合衰老模型大鼠海马中糖皮质激素受体的基因表达,从而减轻慢性应激对大鼠海马的损伤,起到延缓衰老的作用。
Objective: To observe the influence of acupoint catgut embedding on hippocampal glucocorticoid receptor mR- NA expression in aging rat model induced by multi-chronic stress, and to investigate its intervention of aging. Methods: Forty-eight healthy SPF SD rats aged 3 months were randomly divided in 4 groups, blank control group, D-galactose group, multi-aging group, and acupoint catgut embedding group. Blank control group was given intraperitoneal injection with saline for 6 weeks. D-galactose group was given intraperitoneal injection with D-galactose for 6 weeks. Multi-aging group was giv- en intraperitoneal injection with D-galactose for 6 weeks, and from the third week given immobilization stress and noise ex- posure(30 minutes each time) once a day for 4 weeks, and photic-stimulation induced by circadian rhythm inversion. Acupoint catgut embedding group was given the same treatment as the multi-aging group for modeling, and also was given acupoint catgut embedding once per day for 6 weeks. At the end of the experiment, the rats were executed for the examination of hippocampal glucocorticoid receptor mRNA expression. Results: Compared with the blank control group, hippocampal gluco- corticoid receptor mRNA expression was decreased in D-galactose group and multi-aging group, the difference being signifi- cant. Acupoint catgut embedding group had higher expression level of hippocampal glucocorticoid receptor mRNA than the multi-aging group. Conclusion: Multi-aging rats have higher hippocampus glucocorticoid receptor mRNA expression level than D-galactose-inducing aging rats. Acupoint catgut embedding exerts regulatory effect on hippocampal glucocorticoid re- ceptor mRNA expression in multi-aging rats, which results in reducing hippocampus damage induced by chronic stress, and in counteracting aging.
