己酮可可碱对呼吸道合胞病毒所致的TNF-α基因表达水平的影响 被引量:1

摘要 观察己酮可可碱 (PTX)对呼吸道合胞病毒 (RSV)所诱导的巨噬细胞肿瘤坏死因子 (TNF-α)含量及 TNF-α m RNA表达水平的影响。收集 Balb/c小鼠腹腔巨噬细胞 ,随机分成三组 1对照组 (NOR组 ) ;2感染组 (RSV组 ) :培养基中含有 10 6pfu的 RSV;3己酮可可碱组 (PTX组 ) :用 RSV (10 6pfu)感染后 ,培养基中加入 PTX (1m g/ml) ;每组于 RSV感染巨噬细胞 2 0 h后 ,用 EL ISA法测定培养细胞上清中 TNF-α含量的变化 ,同时用逆转录聚合酶链反应(RT- PCR)测定各组巨噬细胞 TNF-α m RNA表达水平。结果表明 :感染组中 TNF-α m RNA表达水平与对照组相比明显增多。 PTX组 TNF-α m RNA的表达和感染组相比明显降低。 TNF-α的含量也有相应的变化 :与对照组比较 ,感染组明显升高 (P<0 .0 1) ;而 PTX组与感染组比较明显下降 (P<0 .0 1)。认为 PTX能抑制 RSV所诱导的巨噬细胞 TNF-α的基因表达 ,并减少 The effect of pentoxifylline (PTX) on level of the TNF α and expression of TNF α mRNA induced by respiratory syncytial virus (RSV) were studied.Peritoneal macrophages were collected from Balb/c mice and randomly divided into three groups: control group (NOR group), RSV infecting group (RSV group), containing 10 6 pfu RSV in the culture medium, and PTX group wiht 1 mg/ml of PTX being added into the culture medium after RSV infection. Twenty h after the RSV infection, the mRNA expression level of TNF α gene in the supernatant was detected by using RT PCR and TNF α protein level by using ELISA method, respectively. The results showed that the mRNA expression level of TNF α gene was increased in the RSV group as compared with that in the NOR group, while it was obviously decreased in PTX group as compared with RSV group. Similar results were also obtained in the changes of TNF α. It was suggested that PTX exerted an inhibitory effect on the expression of the TNF α gene induced by RSVon the level of its mRNA and reduce the production of TNF α protein.
