结肠康对恶唑酮诱导的小鼠溃疡性结肠炎的作用

Effects of Jiechangkang on oxazolone-induced colitis in mice

目的:观察中药复方结肠康对恶唑酮(oxazolone,OXZ)诱导小鼠溃疡性结肠炎(ulcerative colitis,UC)的疗效。方法:小鼠颈背部3%OXZ致敏2 d,第6天用1.5%OXZ灌肠,灌肠当日开始灌胃不同剂量结肠康和阳性对照药柳氮磺胺吡啶(wil-low nitrogen sulfonamides pyridine,SASP),每日记录小鼠体质量,观察小鼠的进食、毛色、粪便、存活等情况,灌胃3 d后处死小鼠,取结肠观察大体形态,根据模型组病变严重部位各组取相同位置固定,常规HE染色,观察各组结肠组织病理学改变。结果:与空白对照组相比,模型组小鼠懒动、精神变差、体质量明显降低、进食减少、毛色无光泽和活动量减少,死亡率高,小鼠疾病活动指数(disease activity index,DAI)明显增高(P<0.01);结肠组织大体损伤评分均明显增高(P<0.01),结肠充血、溃疡、炎症反应较重。然而,结肠康25.6、12.8、6.4 g·kg-1组不同程度地改善OXZ引起小鼠上述异常改变,且作用呈剂量依赖性关系;结肠康25.6 g·kg-1组与SASP0.2 g·kg-1组作用接近。结论:结肠康可明显抑制OXZ诱发小鼠溃疡性结肠炎,减结肠炎症和促进肠道溃疡愈合,对小鼠溃疡性结肠炎具有一定的治疗作用。

ORIECTIVE To observe the effects of Jiechangkang on colon of oxazolone-induced ulcerative colitis （UC） in mice. METHODS The allergy was produced by dropping 3 % oxazolone on micers skin and subsequently with intragastric administration 1.5% oxazolone on 6th day after sensitization. Simultaneously, the mice were given orally three doses of Jiechang- kang. The mice body weight, eating, hair, stool and survival were recorded everyday. The mice were decapitated on the day 3after administration. The pathological changes of colon were observed with microscope Dy HE staining. fonamides pyridine （SASP） was as reference drug. RESULTS Compared with the control group, the model group rats＇ eating was lower, the hair was poorer, the physical activity was fewer and the mortality was higher in model group rats. The disease activity index （DAD and general injury scores in model group rats were also significantly higher than those of control group rats （P〈0. 01）. However, the three doses of 6. 4, 12. 8 and 25.6 g·kg^-1 Jiechangkang differently improved the above abnormal colon changes induced by oxazolone. The reference drug SASP was also found the similar results. CONCLUSION The present Study gets a result that Jiechangkang exerts an improvement on UC mice induced by oxazolone, including colonic pathologicah colonic tissue inflammatory, and intestinal ulcers restored, which suggest that Jiechangkang may have helpful effect in treat ment of UC.
