摘要
应用高效液相色谱法测定鼠骨骼肌细胞ATP、ADP和AMP的含量 ,从离体 (Invitro)和整体(Invivo)两方面分别比较补气药 (人参、黄芪、党参、白术 )与理气药 (枳实、厚朴、青皮、乌药 )对大鼠大腿骨骼肌细胞能荷值的调节作用 ,同时观察补气药所含的多糖类成分对骨骼肌细胞能荷值的影响 (离体 )。结果 :离体和整体实验都表明 ,补气药均升高骨骼肌细胞能荷值 ,而理气药则降低 ;另从人参、黄芪、党参、白术等单味药中 ,用植物化学方法提取分离多糖类成分 ,这些多糖成分均未表现增加骨骼肌细胞能荷值的作用 ,反而有降低的趋势 ,且随浓度的增加呈一定的剂量依赖关系。结论 :补气药与理气药对大鼠大腿骨骼肌细胞能荷值存在明显的差异 ,但未能证实补气药所含的多糖类成分是升高骨骼肌细胞能荷值的物质基础。
Objective - to compare the regulating effects of TCM qi invigorating and qi regulating drugs on the energy charge of the skeletal muscle cells in rat's thigh (SMCRT) both in vivo and in vitro, and to observe the effects of the polysaccharides extracted from qi invigorating drugs on the energy charge of SMCRT in vitro. Method - HPLC was used to determine the contents of ATP, ADP and AMP in SMCRT, and the value of the energy charge was calculated according to a routine formula. In the experiment, the qi invigorating drugs included ginseng, astragalus root, pilose asiabell root, and white atractylodes rhizome; and the qi regulating drugs were immature bitter orange, magnolia bark, green tangerine, and lindera root. Results - Both in vivo and in vitro tests showed that the qi invigorating drugs could increase the value of the energy charge while qi regulating drugs acted on the contrary. The polysaccharides showed no effect of increasing the value of the energy charge, but a tendency to decrease the value in a dose effect manner. Conclusion - qi invigorating and qi regulating drugs acted differently on the energy charge of SMCRT, and it could not be proved that the polysaccharides are the material basis of the qi invigorating drugs in their energy charge increasing action.
