摘要
目的:对致肾盂肾炎大肠杆菌p菌毛的结构基因papA的免疫保护作用进行研究。方法:用PCR方法扩增papA基因,并将554bp扩增产物克隆入真核表达载体pcDNA3,构建重组质粒pCT37作为DNA疫苗,免疫BALB/c小鼠。结果:全菌ELISA测定显示疫苗接种组小鼠血清效价明显高于对照组,分别为1:5750和1:4000(P<0.01)。尿液和肾脏菌落计数在接种组均明显低于对照组(P<0.05),两组动物肾脏病理学检查无明显差别。结论:本研究初步显示了papA良好的免疫保护作用。
Objective:To investigate the immunoprotective effects of papA of Uropathogenic Escherichia coli(UPEC). Methods: A 554bp long DNA fragment encoding PapA mature polypeptide was amplified using Polymerase Chain Reaction(PCR) ,and was cloned into eukaryotic expressing plasmid pcDNA3. The recombinant plasmid pCT37 was employed as DNA vaccine to immunize BALB/c mice. Results:The antibody titers in experiment group(l:5 750) was significantly higher than that in control group(l:4 000)using whole bacterial ELISA(P<0.01).Both urine and renal colony counts indicated significantly less frequently colonization in experiment group(P<0.05),though there was no significant difference in renal invasion between two groups. Conclusion:papA has a good immunoprotective effect.
