十二指肠空肠空置手术对T2DM大鼠胰腺GLP-1受体表达影响的研究 被引量:2

摘要 目的研究十二指肠空肠空置手术(DJB)对2型糖尿病(T2DM)大鼠胰腺胰高血糖素样肽-1受体(GLP-1R)表达的影响,探讨DJB手术后胰腺GLP-1R表达变化与血糖的关系。方法雄性Wistar大鼠随机分为4组,每组8只:Wistar正常大鼠组(W-C);T2DM模型对照组(T2DM-C);T2DM二甲双胍组(T2DM-M);T2DM+DJB手术组(T2DM-DJB)。分别测定各组大鼠术前及术后第2、4、8周空腹血糖、空腹血清GLP-1和空腹血清胰岛素值。术后第8周处死大鼠,RT-PCR检测胰腺GLP-1RmRNA的表达变化。结果 T2DM-DJB、T2DM-M组大鼠术后空腹血糖和空腹血清胰岛素显著降低(P<0.01),空腹血清GLP-1显著升高(P<0.01),与相应时间点T2DM-C组相比均有统计学意义(P<0.01)。T2DM-DJB组大鼠术后空腹血糖、空腹血清GLP-1、空腹血清胰岛素与T2DM-M组大鼠相比无明显差异(P>0.05)。术后第8周,RT-PCR结果显示GLP-1RmRNA在T2DM-DJB、T2DM-M组大鼠胰腺表达水平分别为(0.55±0.03)、(0.53±0.01),明显高于T2DM-C组大鼠GLP-1R mRNA的表达水平(0.41±0.02)(P<0.01)。结论 DJB手术能显著降低T2DM大鼠血糖,可能与术后T2DM大鼠胰腺GLP-1R表达水平升高有关。 Objective To study the effects of duodenal jejunal bypass(DJB) on the expression of glucagon-like peptide-1 receptor(GLP-1R) in the pancreas of type 2 diabetes(T2DM) rats, and explore the relationship between GLP-1R expression and blood glucose. Methods Wistar rats were randomly divided into four groups: Wistar control group(W-C), T2DM control group (T2DM-C),T2DM Metformin treated group (T2DM-M), T2DM DJB surgery group(T2DM- DJB), 8 rats in each group. Fasting blood glucose, fasting insulin and fasting GLP-1 were detected before DJB operation and 2 weeks, 4 weeks, 8 weeks after DJB operation. After 8 weeks, the rats were sacrificed. RT-PCR Results Fasting blood glucose, was used to analysis the mRNA levels of GLP-1R in the pancreas. and fasting insulin of T2DM-DJB and T2DM-M rats decreased after DJB surgery, and fasting serum GLP-1 increased after DJB surgery (P〈0.01). They were sig- nificantly different compared with T2DM-C group in the same time(P〈0.01). Fasting blood glu- cose, fasting insulin and fasting serum GLP-1 of T2DM-DJB were not significantly different com- pared with T2DM-M(P^0.05). After 8 weeks, the expressions of GLP-1R mRNA in T2DM- DJB and T2DM-M pancreas cells were (0.55±0.03) and (0.53±0.01), it also increased signifi- cantly compared with T2DM-C rats (P〈0.01). Conclusion DJB surgery can significantly decrease T2DM rats fasting blood glucose. Maybe the high expression of GLP-1R in pancreas after DJB surgery plays an important role in decreasing the fasting blood glucose of T2DM rats.
