摘要 目的观察环氧化酶-2(COX-2)及肺耐药蛋白(LRP)在非小细胞肺癌(NSCLC)组织中的表达,探讨COX-2介导NSCLC耐药与LRP的关系。方法采用免疫组织化学法,检测58例NSCLC标本与22例正常肺组织中COX-2和LRP的表达水平,并分析其表达与NSCLC患者的年龄、性别、组织学类型、分化程度、淋巴结转移及分期的关系。结果 COX-2与LRP在NSCLC组织中阳性表达率高于正常肺组织(60.3%vs31.8%,75.9%vs41.0%,P均<0.05),在肺腺癌中的表达水平高于鳞癌的表达水平(P<0.05);NSCLC中COX-2与LRP的表达与患者的年龄、性别无明显关系(P均>0.05);而与组织学类型、分化程度、淋巴结转移及分期有关(P均<0.05);NSCLC组织中COX-2与LRP的表达呈正相关(r=0.309,P<0.05)。结论 COX-2及LRP在NSCLC中高表达,COX-2可能通过调控LRP介导NSCLC原发性耐药,检测两者表达水平对肺癌的综合治疗起到一定的指导作用。 Objective To observe the expression of cyclooxygenase-2(COX-2) and lung resistance protein(LRP) in non-small cell lung caneer(NSCLC), and explore the relationship between COX-2-mediated drug resistance and LRP in NSCLC. Methods The expression of COX-2 and LRP was detected by immunohistochemistry in 58 cases NSCLC specimens and 22 cases normal lung tissues. The correlations of COX-2 and LRP expression with age, sex, histological type, differentiation, lymph node metastasis and staging were explored. Results The positive expression rate of COX-2 and LRP in NSCLC was higher than that of normal lung tissues(60. 3X vs31.8%, 75.9%vs41%, P both〈0.05), the expression level in pulmonary adenocareinoma was higher than that of squamous cell carcinoma(P〈0.05) ; Neither COX-2 nor LRP expression was related with patient age and sex(P both〉0.05), but related with histological type, differentia- tion, lymph node metastasis and staging (P a11〈0.05) ; There was a significant positive correla- tion between COX-2 and LRP expression in NSCLC(r=0. 309, P〈0. 05). Conclusion The ex- pression of COX-2 and LRP in NSCLC was significantly higher; COX-2 mediates primary drug resistance in NSCLC perhaps through regulating the expression of LRP. Detecting the expression level of COX-2 and LRP has a certain guiding role in the comprehensive treatment of lung cancer.
