摘要
目的探讨血管生成素-1基因(Ang1)表达对人胃癌细胞增殖与凋亡的影响,为肿瘤靶向治疗策略的设计和选择提供理论依据。方法以感染复数为20的对照病毒Ad-GFP和重组病毒Ad-Ang1感染胃癌细胞,比色法(MTT法)分析Ang1对胃癌细胞增殖的影响,流式细胞仪分析血浆饥饿时对其凋亡的影响。结果 Ad-Ang1组与对照组或Ad-GFP组相比,吸光度(A)值显著增加(P<0.05),说明转染Ang1基因能促进MGC-803细胞的增殖。Ad-Ang1组与对照组或Ad-GFP组相比,凋亡率显著下降(P<0.05),说明转染Ang1基因能抑制血浆饥饿条件下的细胞凋亡。结论胃癌的发生发展是个复杂的过程,Ang1基因转染后能显著促进体外培养的胃癌细胞增殖,并可抵抗血浆饥饿所诱导的细胞凋亡。
Objective To investigate the effects of angiopoietin-1 gene expression on the proliferation and apoptosis of human gastric cancer cells as a theoretical basis for tumor targeting. Methods Virus Ad-GFP and recombinant virus Ad-Angl were used to infect gastric cancer cells (multiplicity of infection = 20). A colorimetric assay (MTT) was used to analyze the effects of Angl on gastric cancer cell proliferation and flow cytometry was used to analyze the effects of Angl on gastric cancer cell apoptosis during plasma deprivation. Results Cells infected with Ad-Angl had a significant- ly higher absorbance compared to those infected with Ad-GFP and the control group (P^0.05), indicating that Angl gene transfection significantly promoted the proliferation of MGC-803 cells. Cells infected with Ad-Angl had a signifi-cantly lower rate of apoptosis compared to cells infected with Ad-GFP and the control group (P〈0.05), indicating that Angl gene transfection significantly inhibited apoptosis during plasma deprivation. Conclusion The incidence of gastric cancer is a complex process. Angl gene transfection significantly promotes the proliferation of gastric cancer cells in vitro and inhibits apoptosis resulting from plasma deprivation.
