摘要
目的:探讨缺氧诱导因子-1α(HIF-1α)和碳酸酐酶-9(CAⅨ)在子宫颈鳞状细胞癌(SCC)组织中的表达及其临床意义。方法:采用EliVision Plus/HRP免疫组织化学法检测HIF-1α和CAⅨ在30例正常子宫颈组织、55例子宫颈上皮内瘤变(CIN)和80例SCC组织中的表达特点及相关性。结果:HIF-1α在正常子宫颈组织、CIN及SCC组织中的表达率分别为0.00%、43.64%和76.25%,差异均有统计学意义(P<0.01);CAⅨ在正常子宫颈组织、CIN及SCC组织中的表达率分别为0.00%、40.00%和66.25%,差异均有统计学意义(P<0.01);低分化SCC组织中HIF-1α、CAⅨ的表达率高于高-中分化SCC组织(P<0.05),有淋巴结转移SCC组织中HIF-1α、CAⅨ的表达率高于无淋巴结转移SCC组织(P<0.05),Ⅱ期SCC组织中HIF-1α的表达率高于Ⅰ期SCC组织(P<0.05);SCC组织中HIF-1α与CAⅨ的表达呈正相关关系(P<0.05)。结论:HIF-1α、CAⅨ在SCC的发生、发展中起重要作用,SCC中HIF-1α和CAⅨ的高表达和正相关关系提示了以缺氧诱导因子通路为基础的分子靶向治疗的可行性。
Objective: To explore the clinical significance and expressions of hypoxia-induced factor-lct (HIF-lct) and carbonic anhydrase IX (CA IX ) in cervical squamous cell carcinoma(SCC). Methods:The expression characteristics and correlations of HIF-1 a and CAIX in normal cervical tissues of 30 cases,cervical intraepithelial neoplasia(CIN) of 55 cases and SCC tissues of 80 cases were detected by immunohistochemistry of EliVision Plus/HRP. Results:The positive expression rates of HIF-la in normal cervical tissues, CIN tissues and SCC tissues were increased gradually, whose were 0.00% ,43.64% and 76.25% , respectively, and their differences were statistical significant( P 〈 O. O1 ). The positive expression rates of CA IX in normal cervical tissues, CIN tissues and SCC tissues were increased gradually, whose were O. 00% ,40.00% and 66.25 % , respectively, and their differences were statistical significant (P 〈 O. O1 ). The expression rates of HIF-la and CAIX in poorly differentiated SCC were significantly higher than those in well or moderated differentiated SCC (P 〈 0.05 ). The expression rates of HIF-1 a and CA IX in SCC with lymph node metastasis were significantly higher than those in SCC without lymph node metastasis ( P 〈 0.05 ). The HIF-1 ct expression rates in SCC of clinical stage II were significantly higher than those in clinical stage I (P 〈 O. 05 ). The expression of HIF-1 ct was significantly related to the CA IX expression in SCC (P 〈 O. 05 ). Conelusions:HIF-1 ct and CAIX play an important role in the occurrence and development of SCC. The high expression and postive correlations of HIF-lct and CA])( in cervical SCC suggest the feasibility of the targeted molecular therapy based on the pathway induced by hypoxia factor for treatment of SCC.
