摘要 观察血小板源性生长因子 (PDGF)对于培养人脐静脉内皮细胞 (HUVEC)增殖的影响 ,为血管再生性治疗的靶基因选择提供线索。用流式细胞仪分析细胞周期各时相分布 ,氚 -亮氨酸 ( 3H -Leu)掺入实验评价蛋白质合成 ,免疫组织化学方法检测增殖细胞核抗原 (PCNA)水平。结果不同浓度PDGF均可加速内皮细胞由G1期→S期的转换 ,使细胞进入G2 /M期 ,增加培养细胞3H -Leu掺入量及PCNA水平 ;但PDGF浓度为 5ng时与对照组相比 ,作用不显著。PDGF能够促进二维培养内皮细胞增生 。 Objective The aims of our studies was to disclose whether platelet derived growth factor could facilitate the proliferation of culture endothelial cell.Methods The proliferation of HUVEC under normal condition was assayed as followed:cell cycle detected by flow cytometry,protein synthesis by observed 3H-Leucine corporation,proliferating cell nuclear antigen (PCNA) by immunohistochemistry.Result HUVEC transform from G 1 phase to S phase can be accelerated by PDGF,the 3H-Leucine corporation and PCNA level of HUVEC can be increased,it was showed dose dependent.The PDGF of 5ng/ml was ineffective and the effect become stronger as the dose increase.Conclusion The present can be accelerated by PDGF,series of studies suggested that HUVEC proliferation can be accelerated by PDGF.
