摘要
采用冷冻干燥结合辐照新技术处理人脐动脉 (简称LIHUA)做为小动脉旁路移植新材料。通过 7条犬建立颈动脉间置移植实验模型 (左颈动脉内衬间皮细胞 ,右侧对照 )。应用动脉造影、光镜和扫描电镜等方法来观察新移植物在体内的组织修复过程及其内衬效果 ,以了解其组织适应性。有许多滋养血管穿过外膜向内延伸 ,术后 6周血管内皮细胞再生良好 ,总通畅率为 71%。虽然内衬间皮细胞的数量和活性细胞的比例尚高 ,但存留率较低 ,内衬结果不满意 ,2组间的组织修复和通畅率无显著差异。血管内皮细胞的再生属于“同步再生”的模式 ,所以内衬间皮细胞技术不适用于LIHUA。说明冻干辐照人脐动脉的组织适应性良好 ,符合同 (或异 )种小口径动脉移植新材料的基本要求。
Lyophilize Irradiated Human Umbilical Artery (LIHUA) is able as a new prosthesis for small caliber artery bypass grafting.Its histocompatibility and the effect of mesotheliel cell suspension on the patency of LIHUA were carried out in 7 dogs.A carotid artery model was established.Each dog was divided into two groups and received arterial interposition for one seeded (left) and another nonseeded (right) or controlled LIHUA.The prosthesis was explanted at 2,4,6,7,8,9 and 10 weeks after replacement.It were assessed by angiography,light and scanning electron microscopy.A lot of vasa vasorum could be found on this evidence that pass through adventitia into media.Six weeks postoperatively,endothelial cells were well developed that was approved by scanning electron microscope.The total patency rate of the grafts was 71%.Although the number of harvested mesothelial cell and percentage of viable cell were rather high,yet the seeding density and its retaining rate were low.It is therefore,the seeding procedure was unsuccessfully.There was no significant effect of seeded group on patency and histological change.It is suggested that the spontaneous endothelization was established with a model of “simultaneous regeneration”.New endothelium may be regenerated at the same time both on the graft and anastomotic site.Therefore,cell seeding procrdure would not be considered in this situation.In our study,the healing peocess of the patent prosthesis,LIHUA,was satisfied,and its good histocompatibility was also approved.
