链脲佐菌素诱导的大鼠海马神经元超微结构变化 被引量:1

摘要 目的探讨与糖尿病相关的老年性痴呆(Alzheimer’s disease,AD)急性动物模型脑内超微结构变化。方法利用糖尿病模型的诱导药物链脲佐菌素(streptozcin,STZ),通过其侧脑室注射也同样能够诱导大鼠AD样病理变化。并研究通过投射电子显微镜(transmission electronic microscope,TEM)观察STZ侧脑室注射后大鼠海马神经元超微病理学改变。结果STZ的急性侧脑室注射诱导了大鼠海马神经元一系列的超微结构变化,其中包括细胞核变化、内质网异常等,并且发现了突触囊泡和核蛋白体的异常聚集。结论 STZ侧脑室注射诱导的大鼠神经元超微结构异常证实了糖尿病与AD之间特殊的病例关系,为将AD作为一种脑内代谢性疾病进行防治提供了一定的实验室基础。 Objective To explore the ultrastructure changes of Alzheimer's disease(AD) animal model relative with diabetes.Methods STZ was used to induce the AD model in rats though intracerebroventricular injection(ICV).And the ultrastructure changes of the neurons in hippocampus of the rats were observed.Results The short-term ICV of STZ induced a series of ultrastructure changes of the neurons in the hippocampus of rats including alternative nuclear structure,endoplasmic reticulum(ER) abnormality etc.Besides,the abnormalities of synaptic vesicles and ribosomes were observed.Conclusion The alternations of the ultrastructure changes induced by ICV of STZ prove the unique relationship between AD and diabetes,which provides a foundation for making an anti-diabetic agent as a novel therapeutic drug to AD.
