CD44干扰对人宫颈癌HeLa细胞体外转移的影响及其作用机制

摘要 目的研究CD44表达降低对人宫颈癌HeLa细胞体外转移能力的影响及其作用机制。方法用特异性shRNA对HeLa细胞内的CD44进行干扰,采用细胞划痕试验和Transwell侵袭试验分别检测HeLa细胞的迁移和侵袭能力,Real-time RT-PCR、Western blot法分别检测HeLa细胞膜型基质金属蛋白酶1(MT1-MMP)mRNA及蛋白的表达。结果 HeLa细胞内CD44表达降低后,HeLa细胞损伤愈合速度降低,细胞穿过滤膜的数量减少,MT1-MMPmRNA和蛋白表达量降低(P均<0.05)。结论 CD44表达降低可有效抑制HeLa细胞的体外转移能力,其作用机制可能与降低MT1-MMP表达有关。
