丝裂霉素C对大鼠硬膜外瘢痕中转化生长因子β1表达的影响 被引量:1

摘要 目的观察丝裂霉素C对大鼠椎板切除术后硬膜外瘢痕中转化生长因子β1(TGF-β1)表达的影响及探讨其预防硬膜外瘢痕粘连的机制。方法 40只成年大鼠随机分组,每组10只,手术切除L1及L2椎板,根据不同组别以棉片浸透生理盐水或不同浓度的丝裂霉素C置于裸露的硬脊膜后方5 min,2周后取椎板缺损区组织切片做HE染色、免疫组化染色及计算机图像分析。结果 2周时对照组的硬膜外瘢痕形成明显,而实验组中随着丝裂霉素C浓度的增加,硬膜外瘢痕明显减少,实验组TGF-β1免疫组化染色阳性细胞数明显少于对照组。结论丝裂霉素C能抑制大鼠硬膜外瘢痕中TGF-β1的表达,抑制成纤维细胞增殖,降低硬膜外瘢痕的形成。 Objective To observe the effect of mitomycin C (MMC) on the expression of the TGF-β1 in the epidural scar after laminectomy and to clarify the mechanism of MMC for preventing epidural scar after laminectomy. Methods Laminectomy was performed at L1 and L2 in 480 rats, then the rats were divided into 4 groups: high (H), middle (M), low (L) and control (C), soaked with different concentrations of MMC (0.05, 0.1, 0.5 g/L) or saline on the laminectomy defects for 5 min, respectively. After two weeks, the laminas of defect area were taken for HE staining, immunohistochemical staining and computer-assisted image analysis. Results Gross evaluation: there were statistic differences in the grades of adhesion between the control groups and experimental groups, P 〈0.05 (rank sumtest,P=0.035). Conclusion MMC can reduce the expression of TGF-β1 in epidural scar. The possible mechanism of preventing epidural scar after laminectomy by use of MMC could be reducing hematoma and expression of TGF-β1 in epidural scar.
