摘要
目的探讨支气管哮喘患者外周血T细胞OX40及血清可溶性OX40配体(sOX40L)的表达及意义。方法流式细胞术检测65例支气管哮喘患者(发作期哮喘、缓解期哮喘)及30例健康志愿者(对照组)外周血T细胞表面OX40的表达水平,ELISA检测血清sOX40L的表达量,并测定用力1秒呼气率(FEV1%)、呼出气一氧化氮(FeNO)、外周血嗜酸性粒细胞(EOS)计数、总免疫球蛋白E(IgE)及哮喘控制测试问卷(ACT)评分。结果发作期哮喘组患者外周血CD4+T细胞上OX40表达水平及血清sOX40L含量均显著高于缓解期患者和对照组(P<0.05)。相关分析显示,发作期哮喘组OX40表达与FeNO表达、EOS和IgE呈正相关;血清sOX40L表达与FeNO和EOS呈正相关,与FEV1%呈负相关。结论 OX40及血清sOX40L在哮喘患者外周血中异常增加,可作为反映支气管哮喘的炎性浸润程度和疾病严重程度的生物学指标。
Objective To investigate the expression of co-stimulatory molecule OX40 on T-cells and serum concentration of soluble OX40 ligand (sOX40L) in asthmatic patients. Methods The expression of OX40 on T lymphocytes in peripheral mononuclear cells(PBMCs) was detected by flow cytometry in 65 asthmatic cases on attack(group A1) or in ease stage(group A2) and 30 healthy people(group C). Serum sOX40L was determined by ELISA. The ratio of forced expiratory volume in 1 second to vital capacity(FEV1 ~ ), eosinophils (EOS), IgE and expiratory nitric oxide (FeNO) were tested as well. The ACT score was recorded. Results Compared with group A2 and group C, the expression level of CD4+ OX40+ and serum concentration of sOX40L in group A1 were significantly higher(P〈0. 05). The expression level of CIM+OX40+ was positively correlated to the FeNO, EOS and IgE, but negatively correlateed with FEV1 ~ in group A1. Conclusion Abnormally increases of OX40 and sOX40L exist in asthmatic patients, which may be taken as the meaningful biomarkers reflecting the inflammatory status and the severity of bronchial asthma.
