中风膏对动脉粥样硬化模型家兔血脂水平、血管内皮功能的影响 被引量:7

摘要 目的:探讨中风膏对动脉粥样硬化(atherosclerosis,AS)家兔模型的作用机制。方法:将50只家兔随机分为空白对照组、动脉粥样硬化模型组(AS组),中风膏低、高剂量组,辛伐他汀组。除空白对照组外其余各组采用高胆固醇饲料喂养8周制备颈动脉粥样硬化家兔模型。通过全自动生化分析仪检测血清TC、TG、LDL-c含量;分光光度计检测血清NO含量、放射免疫法检测血清ET-1含量;颈动脉组织切片HE染色,光镜观察动脉粥样硬化形态学。结果:AS组家兔血清TC、TG、LDL-c含量明显高于空白对照组(P<0.05)。病理形态学显示AS组颈动脉壁多处出现典型粥样斑块,而空白对照组无异常变化,表明动脉粥样硬化模型复制成功;中风膏高、低剂量组,辛伐他汀组血清TG、TC、LDL-c、ET-1含量均降低,NO升高,与空白对照组相比有明显差异(P<0.05)。中风膏高剂量组、辛伐他汀组中动脉粥样硬化家兔模型的斑块形态、斑块面积均较空白对照组和中风膏小剂量组明显改善(P<0.05)。结论:中风膏能明显降低AS家兔模型血清TC、TG、HDL-c、ET-1水平,升高NO水平,从而达到保护血管内皮细胞、抗动脉粥样硬化的作用。 Objective: To explore the mechanism of ZhongFengGao on rabbits with atherosclerosis (AS). Method: All 50 rabbits were randomized into blank control group, model group, low and high dose groups of ZhongFengGao groups as well as treatment group of simvastatin. Except blank control group, other groups were fed with high cholesterol food to build the models with AS. Contents of TC, TG and LDL-c in the serum were detected with automatic biochemical analyzer; NO content was measured with spectrophotometer and ET-1 content with radioimmunoassay; tissue slice of carotid artery was stained with HE and observed under light microscope. Result: Contents ofTC, TG and LDL-C in the serum of AS group were higher than that of the control group (P〈0.05). Pathomorphology suggested that there were typical atheromatous plaque appeared on paries carotica of AS group, there was no abnormality in the control group, which meant that the models were successfully copied. Contents of TC, TG and LDL-C in the serum decreased but NO rose in low and high dose groups of ZhongFengGao groups and treatment group of simvastatin, compared with the control group, it showed significant difference (P〈0.05). Shape and areas of plaque in rabbits with AS of high dose group of ZhongFengGao and treatment group of simvastatin were improved greater than that of the control group and low dose group of ZhongFengGao (P〈0.05). Conclusion: ZhongFengGao could obviously lower the levels of TC, TG, HDL-c and ET-1, raise NO level to protect vascular endothelial cell and against AS.
