摘要
为了探索丙型肝炎病毒核心区基因疫苗对小白鼠细胞及体液免疫功能的影响,本文将HCV 核心区基因定向克隆于质粒pcDNA3 巨细胞病毒启动子下游,构建了能在真核细胞内表达的重组基因疫苗pcDNA3-HCV,将此疫苗通过多点肌肉注射免疫BALB/C 小鼠,结果发现接种了该疫苗的小鼠出现抗-HCV IgG 阳性,其抗体水平明显高于对照组,同时Th1 免疫应答相关的细胞因子IL-2 及g 干扰素水平也明显高于对照组,其结果表明,HCV 核心基因疫苗能诱导BACB/L 小鼠产生良好的细胞及体液免疫应答。
The aim of the study is to know that HCV DNA vaccine could induce the cellular and humoral immunity in mice or not.The pcDNA3-HCV in which the core gene sequences of HCV were inserted into downstream of CMV promoter in pcDNA3 were injected intramuscularly into BALB/C mice The results showed that the serum anti-HCV were detected in all of the pcDNA3-HCV immunized mice but none in control mice TH1 cytokines such as interleukine 2 and interferon gamma were significantly higher in pcDNA3-HCV immunized mice than that in controls It was indicated that the good responses of cellular and humoral immunity can be induced by injection of pcDNA3-HCV in BACB/C mice
