摘要
目的探讨胰岛素样生长因子1(IGF-1)与肾间质纤维化的关系。方法采用单侧输尿管结扎致肾间质纤动物模型,将30只Wistar雄性大鼠,随机平均分为假手术组、模型组和治疗组,术后14 d,测肾功能,免疫组织化学、原位杂交法,测Ⅳ型胶原蛋白、肌动蛋白α(α-SMA)、组织蛋白酶抑制剂1/基质金属蛋白酶9 mRNA的表达。结果模型组、治疗组血尿素氮、血肌酐,Ⅳ型胶原蛋白、α-SMA、组织蛋白酶抑制剂1水平高于假手术组(P<0.05);而治疗组与模型组相比MMP-9水平显著降低(P<0.05)。结论 IGF-1参与梗阻性肾病的病理生理过程,导致间质纤维化和肾功能损害。
Objective To investigate the effects and mechanisms of IGF-1 in rats with unilatreal ureteral obstruction(UUO).Methods 30 UUO Wistar male rats were randomly divided into pseudo-operation group,model group and treatment group.14 days after the operation,the renal function was tested.The expression of col-Ⅳ,α-SMA,TIMP-1/MMP-9mRNA in kidneys were observed by immunohistochemistry and in situ hybridization.Results BUN,Ser col-Ⅳ,α-SMA,TIMP-1 mRNA of the model group and treatment group were higher than the pseudo-operation group(P0.05);MMP-9 of the treatment group was significantly lower than the model group(P0.05).Conclusion IGF-1 participates in the pathophysiological process,resulting in the renal interstitial fibrosis and renal function impairment.
