摘要
目的:观察迷迭香酸对大鼠肾草酸钙结石形成和间α胰蛋白酶抑制物(IαI)表达的影响。方法:用乙二醇和氯化铵灌胃大鼠造成草酸钙结石模型,应用迷迭香酸不同剂量作为干预因素,肾石通颗粒剂作为阳性对照药。4周后测定尿、血生化,观察肾组织病理变化及Von-Kossa染色肾草酸钙结晶形态分布,应用免疫组织化学和计算机图像分析技术检测IαI在肾组织中的表达。结果:迷迭香酸中剂量与高剂量组大鼠的肾小管上皮细胞IαI的表达,24h尿草酸、Ca2+的排泄量及血尿素氮、肌酐、尿酸含量,均显著低于模型对照组;迷迭香酸中、高剂量组肾组织病理变化、草酸钙结晶分布情况与模型对照组比较也明显减轻。结论:迷迭香酸可能通过抑制肾组织内草酸钙晶体的生成和减少肾组织IαI的表达,从而抑制肾结石的形成,对大鼠草酸钙结石有明显防治作用。
Objective: To investigate the effect of rosmarinic acid on renal stone formation and the expression of inter-alpha- trypsin inhibitor (IαI) in rat. Methods: The model control rats with calcium oxalate renal stone formation were induced by ethylene glycol and ammonium chloride for 28 days. And, different doses of rosmarinic acid were applied as the interference factor, and Shenshitong Keli as positive control drug. The contents of urine oxalic acid and Ca2+ and the blood biochemistry af- ter 28d of rats in each group were detected. Von-Kossa's staining was employed to observe renal calcium oxalate crystal morphology distribution and the renal pathological changes. Computer image analysis was carried out to evaluate the immunohistochemical expression of IαI in the rat kidney tissues. Results: There were significant differences in the expression of IαI in the kidney tubules epithelial cells, the oxalic acid, Ca2+ content in urine and BUN, UA, Cr content between the rosmarinic acid middle-dose (0. 05 g/kg) and high-dose groups (0. 1 g/kg) and the renal stone control group. They both had difference in calcium oxalate crystal morphology distribution compared to the renal stone group. Conclusion : Rosmarinic acid can inhibit calcium oxalate renal stone formation and reduce the expression of IαI in rats,
