摘要
目的:初步探讨淫羊藿苷(ICA)干预下亚硝基谷胱甘肽还原酶(GSNOR)及B细胞淋巴瘤/白血病-2基因(Bcl-2)在糖尿病大鼠睾丸中表达。方法:链脲佐菌素建立糖尿病大鼠模型,低、中、高(30、 70、 100mg/kg)淫羊藿苷灌胃21d,光镜观察大鼠睾丸形态学变化,检测外周血血糖、雌二醇及睾酮水平。免疫组织化学检测GSNOR及Bcl2在睾丸中的表达。结果:中剂量组血糖明显降低,低、中剂量组睾酮与雌二醇比值升高;GSNOR主要表达在精母细胞及精子细胞,与对照组相比,低、中剂量组灰度值较高。Bcl-2蛋白主要表达在睾丸间质细胞,与对照组相比,低、中剂量组阳性细胞数明显增多。结论:淫羊藿苷可稳定糖尿病大鼠血糖,提高睾酮与雌二醇比值对睾丸有保护作用,其作用可能与减少睾丸间质细胞凋亡,提高Bcl-2及GSNOR蛋白表达,抵抗亚硝化应激有关。
Objective:To observe the effect of icariin on s-nitrosoglutathione reductase (GSNOR) and Bcl-2 in diabetic rats tes- tis. Methods: Sprague-Dawley rats were induced by an injection of streptozotocin. Four weeks after injection of STZ, the rats were randomly divided into 5 groups: a normal group, a control group, a low dose group (30 mg/kg, ig), a middle dose group (70 mg/kg, ig) and a high dose group (100 mg/kg, ig). The blood of the heart was tested for testosterone and estradiol. Im- munohistochemical technique was used to detect the expression of GSNOR and Bcl-2. Results: Compared with control group, the ratio of testosterone and estradiol in the middle dose group were increased, and the fasting food glucose was descend evi- dently between the control group and the middle dose group. Numerous pathological changes (decreased spermatogenic cell, distortion seminiferous tubule) were lessening in diabetic rats after receiving ICA. Immunohistochemistry showed that there were higher GSNOR protein expressions in the low dose group and middle dose group. A high expression of Bcl-2 protein was observed in Leydig cells. Significant differences existed in the control group, the low dose group and the middle dose group. Conclusion: Icariin evidently relieves testicular damage in diabetic rats by improving the ratio of testosterone and estradiol, reducing the level of FFG and increased the expression of GSNOR and Bcl-2 protein.
