摘要
目的:研究急性高眼压下大鼠视网膜缺氧诱导因子的表达变化;酸枣仁皂甙A对急性高眼压大鼠视网膜缺氧诱导因子(HIF-1α)的影响。方法:建立大鼠急性高眼压模型。造模后,每天给药1次至取材,对模型给药组和对照给药组分别从大鼠尾静脉按20mg/kg剂量注射酸枣仁皂甙A。各组分别于造模后6、 24、 48、 72h取材,H-E染色观察各组视网膜的结构变化;RT-PCR检测HIF-1α mRNA在各组视网膜上的表达。结果:H-E染色显示,造模后,随着时间延长,视网膜神经节细胞排列紊乱,神经纤维层水肿,内界膜破裂不连续;注射酸枣仁皂甙A后,视网膜的结构变化有所改善。RT-PCR检测视网膜上的HIF-1α mRNA表达:各模型组随着造模后时间的延长,HIF-1α mRNA均值逐渐升高,并在48h达到峰值,与空白对照组和空白给药组比较差异有统计学意义;模型给药组的HIF-1α mRNA均值比模型组低,差异有统计学意义。结论:HIF-1α是急性高眼压内早期变化的因子;酸枣仁皂甙A可改善急性高眼压大鼠视网膜的结构;酸枣仁皂甙A可降低24、 48、 72h急性高眼压大鼠视网膜上HIF-1α的表达。
Objective:To observe changes of the expression of hypoxia inducible factor in the retina of model rats with acute in- traocular hypertension, and to study influence of jujuboside A on the expression of the hypoxia inducible factor in the retina of model rat with acute intraocular hypertension. Methods: The anterior chambers of rats were perfused with saline to cause the acute intraoeular hypertension as a model. After modeling, the rats in the model group and the control group were injected im- mediately with jujuboside A by 20 mg/kg dose v/a the tail vein of rats, once a day at 9 clock. Each group was derived accord- ing to the detection method as following: HE staining and structural change observation; observation on the volume of relative expression changes of HIF-1α mRNA in retinal in each group after blank administration. Results: Along the extended time points after modeling, HE staining showed disordered arrangement of retinal ganglion cells, nerve fibrous coat edema, and internal limiting membrane were ruptured and not continuous. Along the extended time points after modeling, the mean of the HIF-1α mRNA relative expression gradually increased and peaked at 48 h, and the difference between the blank and blank treatment groups was statistically significant; the model treatment group had a lower average of the relative expression of HIF-1α mRNA than did that in the model group, and the difference was statistically significant. Conclusion: HIF-1α expressed earlier in the retina of rats with acute intraocular hypertension. Jujuboside A can improve retinal structure with acute intraocular hypertension, but has no effect on the normal retina. Jujuboside A can reduce the expression of HIF-1α at 24 h, 48 h and 72 h in the retina of rats which has acute intraoeular hypertension.
