摘要
目的研究转化生长因子(TGF)-β1和白介素(IL)-6对体外培养CD4+CD25-T细胞表型及细胞因子分泌的影响。方法采用免疫磁珠分选法从健康人外周血中分离CD4+CD25-T细胞和CD4+CD25+nTreg细胞,分别或同时使用TGF-β1和IL-6体外刺激CD4+CD25-T细胞,培养120 h后检测细胞表型,以及细胞培养液中细胞因子水平。结果 CD4+CD25-T细胞在TGF-β1刺激下CD4+CD25+T细胞比例增高,细胞培养液中IL-10、IFN-γ、IL-4升高,IL-17降低,与对照组比较差异有统计学意义(P<0.001);TGF-β1和IL-6联合刺激CD4+CD25-T细胞时,CD4+CD25+T细胞比例减少,细胞培养液中IL-10、IFN-γ、IL-4降低,IL-17水平增高,与对照组比较差异有统计学意义(P<0.001)。结论 TGF-β1可诱导CD4+CD25-T细胞分化为CD4+CD25+Treg细胞;IL-6可抑制TGF-β1诱导CD4+CD25-T细胞分化为CD4+CD25+iTreg细胞,而促进Th17细胞的产生。
Objective To investigate the effects of TGF-β1 and IL-6 on CD4+CD25-T cells phenotype,secretion and functions of cytokine.Methods CD4+CD25-T cells and CD4+CD25+ nTreg cells were isolated with magnetic cell sorting from healthy human peripheral blood.CD4+CD25-T cells were stimulated by TGF-β1 or IL-6 alone or in the combination.The cell phenotype and cytokines were detected in the supernatant after 120 hours.Results While CD4+CD25-T cells were stimulated by TGF-β1,CD4+CD25+ T cell proportion and levels of IL-10,IFN-γ and IL-4 were significantly increased,and IL-17 level was significantly reduced(P0.001).While CD4+CD25-T cells were stimulated by TGF-β1 and IL-6 in combination,CD4+CD25+ T cell proportion was significantly reduced.Levels of IL-10,IFN-γ and IL-4 were significantly reduced,and IL-17 level was significantly increased(P0.001).Conclusions TGF-β1 can induce CD4+CD25-T cells into CD4+CD25+ iTreg cells.IL-6 can suppress this inducing effect of TGF-β1 and promote the generation of Th17 cells.
