大鼠骨骼肌失神经支配后串珠素和突触素表达的变化

摘要 目的:研究串珠素在骨骼肌失神经支配后的表达和神经肌接头的变化。方法:成年SD大鼠30只,左侧坐骨神经切断,右侧正常对照。术后1、2、4周取材。免疫组化检测腓肠肌中串珠素和突触素表达,图像分析表达水平。结果:所有受试大鼠坐骨神经切断后相关肌肉进行性萎缩。神经切断4周后骨骼肌串珠素的表达显著降低(P<0.05),突触素表达也下降,但差异无统计学意义。结论:串珠素在骨骼肌失神经后的表达变化比突触素更明显。 Objective: To study the expression of perleean and synaptophysin (SYN) in the denervated skeletal muscle. Methods: 30 adult rats were equally divided into 3 groups and the left sciatic nerves were cut. The right sciatic nerves were kept as normal con- trois. The rats in the 3 groups were respectively sacrificed 1, 2 and 4 weeks after surgery. The samples of gastrocnemius muscle were prepared for the examination of perlecan and SYN expressions by immunohistochemistry and image analysis. Results: Progressive atro- phy of gastrocnemius was found in all the animals after neuroectomy. Perlecan expression was reduced after neuroectomy ( P 〈 0.05 ). SYN expression was decreased but showed no statistical significancy. Conclusion: Perlecan is more sensitive to the muscular denerva- tion than SYN.
