摘要
比较三种测定血液胆碱酯酶 (ChE)活性的方法。 [方法 ]用改良Ellman法 (A法 )、携带式ChE测定仪法 (B法 )和纸片法 (C法 )同时测定 74名健康人和急性农药中毒患者 5 8人次血液ChE活性 ,统计和分析测定结果。 [结果 ]建立了三种方法血液ChE活性的正常参比值 ,急性氨基甲酸酯、有机磷及有机磷与其他农药混配中毒患者血液ChE活性均有不同程度下降 ,A法中红细胞AChE、全血和血浆ChE以及C法中全血ChE活性百分值与B法中红细胞AChE活性均呈密切的正相关关系 ,相关系数r分别为 0 92 8、0 939、0 6 83和0 90 6。 [结论 ]三种方法均可用于急性有机磷中毒患者ChE活性的测定 ,三种方法各有所长 ,各有所短。
Objective] To compare three methods for measuring blood cholinesterase (ChE) activity. [Method] Blood ChE activity was determined in 132 samples taken from 74 healthy adults non exposed to ChE inhibitor insecticides and 20 intoxicated patients by three different methods. Method A was the improved Ellman spectrophotometry adequate for measuring whole blood,erythrocyte and plasma ChE. Method B was used for measuring erythrocyte acetylcholinesterase (AChE) by a spectrophotometric field kit (Test Mate OP). Method C was used acetylbromocholine as the substrate for measuring whole blood ChE by a test paper. [Results] The reference values of blood ChE activity were established after measuring the blood samples from subjects non exposed to insecticide by three methods. The blood ChE activity was inhibited in 18 cases of organophosphate or its mixed insecticides intoxication and in one case of carbamate intoxication by any method. The activities of erythrocyte AChE,whole blood and plasma ChE measured by method A,and whole blood ChE measured by method C were all significantly correlated with the activities of erythrocyte AChE measured by method B ( r =0 928,0 939,0 638 and 0 906, P <0 01). [Conclusion] All three methods can be used for measuring inhibition of blood ChE activity in patients of organophosphate intoxication. Each has its own strong and weak points.
