摘要
目的研究微环境蛋白TAG-1与神经干细胞分化之间的关系。方法使用免疫荧光染色检测神经干细胞中巢蛋白nestin的表达,并过表达TAG-1后使用实时荧光定量PCR技术分析神经干细胞中分化相关蛋白的表达变化。结果体外培养的神经干细胞中过表达TAG-1以后,干性相关基因nestin和转录因子SOX2(SRY-box2)表达降低,神经元标志物神经元微管蛋白βⅢ(neuronal classⅢβ-Tubulin,Tuj1)和胶质细胞标志物胶质纤维酸性蛋白(GFAP)表达升高。结论 TAG-1可能与神经干细胞的分化存在正相关的关系,其分子调节机制有待进一步研究。
Objective To investigate the association between TAG-1 expression and neural stem cells differentiation. Methods Immunofluorescence stain for nestin was performed to indentify NSCs cultured in vitro. Expression change of differentiation associated genes in mRNA level was detected by Real time PCR. Results After NSCs over expressed TAG-l, the mRNA level of nestin and SOX2 were decreased, Neuronal Class Ⅲ β-Tubulin (Tujl) and glial fibrillary acidic protein (GFAP) were upregulated. Conclusion TAG-1 may participate in the differentiation of neural stem cells, the molecular mechanism needs further research.
