云烟97巨型变异烟株的组织培养与快速繁殖 被引量:3

摘要 为优化云烟97变异烟株组培快繁体系,以嫩叶为外植体,对植物激素的种类、浓度、配比、对外植体切块时间和植物激素添加时间等进行了研究。结果表明,最佳愈伤组织诱导和不定芽分化的培养基配方为MS+6-BA2mg L-1+NAA0.2mg L-1,最佳生根培养基配方为1/2MS+6-BA 0.10 mg L-1+IBA 0.5mg L-1,从转接外植体到组培苗形成需要80 d左右。同时,结果发现,以形成丛芽15~20 d左右切块,有效丛芽数较多、健壮易分割,而激素在培养基灭菌前、后加入对激素的活性影响不大。 Tissue culture and rapid propagation of a Yunyan 97 giant variation plant was carried out. Result indicated that the most suitable medium for callus induction and proliferation of bud clusters was MS+6-BA2mg.L-1+NAA0.2mg.L-1, while the suitable medium for buds amplification was 1/2MS+6-BA 0.10 mg.LI+IBA 0.5mg.L-1. It took about 80 days from explants inoculated to small seedling. Attention was also paid to slicing time of explants with bud clusters and how phytohormone was added. Result suggested that, to slice explants 15 to 20 days after bud spots were formed could promote healthy bud clusters which were also easy to slice. Phytohormone added before and after medium sterilization resulted little influence to phytohormone activity.
