喉鳞状细胞癌中MAD2的表达及临床意义 被引量:1

Expression and Clinical Significance of MAD2 in Laryngeal Squamous Cell Carcinoma

摘要 目的探讨喉鳞状细胞癌(LSCC)组织中有丝分裂阻滞缺陷蛋白(MAD2)的表达及临床意义。方法采用免疫组织化学EliVision二步法检测55例LSCC组织、30例癌旁正常黏膜组织中MAD2蛋白的表达并分析其与LSCC临床病理特征的关系。结果 MAD2蛋白在LSCC组织中的阳性表达率为58.2%(32/55),低于癌旁正常黏膜组织86.7%(26/30),两者差异有统计学意义(P<0.01);中、低分化组MAD2阳性表达率为47.1%(16/34),低于高分化组76.2%(16/21),淋巴结转移组MAD2阳性表达率为35.0%(7/20),低于无淋巴结转移组71.4%(25/35),两组差异均有统计学意义(P均<0.05);MAD2阳性表达与患者性别、年龄、吸烟、肿瘤原发部位、T分期及临床分期无相关性(P均>0.05)。结论 MAD2蛋白在LSCC的发生、分化及转移中可起重要作用,可作为LSCC诊断及预后的参考指标之一。 Objective To investigate the expression and clinical significance of mitotic arrest deficient 2(MAD2) protein in laryngeal squamous cell carcinoma(LSCC) tissues.Methods The expression of MAD2 protein was measured by immunohistochemistry(EliVision two steps) in 55 cases of LSCC tissues,30 cases of adjacent normal laryngeal mucosa tissues,and the relationship of MAD2 expression with clinical pathologic factors in LSCC was also analyzed.Results The positive rates of MAD2 protein in LSCC tissues(58.2%,32/55) was lower than those in adjacent normal laryngeal mucosa tissues(86.7%,26/30),P0.01.Poorly differentiated tumors(47.1%,16/34) expressed lower MAD2 protein to well differentiated ones(76.2%,16/21),P0.05.Tumors with lymph node metastasis(35.0%,7/20) had lower MAD2 protein expression than those without metastasis(71.4%,25/35),P0.01.MAD2 protein expression had no relation to the patient's sex,age,smoking,tumor site,T-stage and clinical stage(P0.05,respectively).Conclusion MAD2 protein may play an important role in the tumorigenesis,differentiation,lymphatic metastasis of LSCC,and can be a useful marker for diagnosis and prognosis of LSCC.
