摘要
构建新的血管内皮生长因子(VEGF)高效真核表达载体pcD 2 /VEGF, 体外转染VSMC 发现其在VSMC中的表达效率较 pcDNA 3 /VEGF 明显增高,体外和体内实验证实VEGF 基因具有显著促进血管生成的作用。
To study the expression and its angiogenesis effect of VEGF gene The human VEGF cDNA was cloned into eukaryotic expression vector pcD2 The recombinant plasmids were transfected into VSMC and bygene suture transferred into the adductor of rats of which the distal extreme of external iliac artery weere ligated With ELISA immunohistochemistry and alkaline phosphotase staining we investigated the in vitro and in vivo expression and its angiogenesis effect of VEGF gene The expression of VEGF gene in VSMC showed that pcD2/VEGF Was significantly higher than pcDNA3/VEGF(1103.86 59.49 vs 751.94 208.13 pg/m1/2 105 P 0.05)In vivo experi- ment also showed that the expression of VEGF gene in rats was evident Fourteen days after the initiation of gene therapy histologically identifiable capillaries were increased in VEGF transfectants compared with controls (16.47 0.33 /10 40 P 0.01).The results demonstrated that the ttansfer of VEGF gene stimulated the formation of focal microvessels and pcD 2 /VEGF was better than pcDNA3/VEGF plasmid for VEGF gene therapy
