摘要
目的通过动物实验于体外条件下观察补肾中药对大鼠骨髓基质干细胞增殖分化的影响。方法将不同浓度的补肾中药方剂与骨髓基质干细胞体外共同培养,测定细胞增殖功能,检测成骨细胞增殖与分化指标(包括碱性磷酸酶、矿化结节)。结果与空白组比较,浓度100 mg/L、200 mg/L的补肾中药在不同时间段均可提高骨髓基质干细胞增殖率(P<0.05);与空白组比较,浓度20 mg/L、40 mg/L、100 mg/L、200 mg/L的补肾中药可使ALP活性增强、矿化结节数量增多(P<0.05)。结论较高浓度的补肾中药能促进骨髓基质干细胞的成骨潜能。
Objective To study the effects of kidney-reinforcing herbal medicine on the differentiation and proliferation of osteoblasts under in vitro conditions. Methods Culture the MSCs and the kidney-reinforcing herbal medicine respectively at the concentrations of 0 mg/L, 1 mg/L, 10 rag/L,20 mg/L,40 mg/L, 100 mg/L,200 mg/L in vitro ;and then evaluation cells proliferation function and detect osoteoblasts indicators such as activity of alkaline phosphatase (ALP) and the mineral node number. Results There is a significant difference in the cell proliferation rate between the high concentrations (100 mg/L, 200 mg/L) with the blank group at any time (P 〈 0.05); the concentrations of 20 mg/L, 40 mg/L, 100 mg/L,200 mg/L groups can elevate ALP activity and improve the mineral node number. Conclusion The higher level of kidney-reinforcing herbal medicine can promote the osteogenic differentiation potential of MSCs.
