The expression and clinical significance of HIF-1α and STAT3 in gastric cancer

摘要 目的研究胃癌中的HIF-1α和STAT3的表达及相互关系,初步探讨他们在胃癌中的生物学意义。方法用免疫组织化学染色(IHC)分别检测60例胃癌组织、15例胃正常黏膜中HIF-1α和STAT3的表达水平,了解HIF-1α和STAT3表达与临床病理指标的关系,以及两者表达的相关性。结果 HIF-1α和STAT3在胃癌组织中的表达均明显高于癌旁正常组织(均P<0.05)。HIF-1α表达与患者性别、年龄及肿瘤部位差异均无统计学意义(均P>0.05),与浸润深度、TNM分期、淋巴结转移相关(均P<0.05)。STAT3蛋白表达与患者性别、年龄及肿瘤部位,差异均无统计学意义(均P>0.05),STAT3与浸润深度、TNM分期、淋巴结转移相关(均P<0.05)。HIF-1α和STAT3表达呈正相关。结论 HIF-1α和STAT3的表达与胃癌生物学行为密切相关;HIF-1α和STAT3可能联合通过抑制靶PTEN基因而促进癌细胞侵袭和迁移。 Objective To study the expression of HIF-lα and STAT3 in gastric cancer, and its corre- lation with the progression of gastric cancer(GC). Methods Expressions of gastric were detected immunohis- tochemistry (IHC) in 30 samples of GC tissue and 10 samples of normal gastric tissue, respectively. The expres- sion of HIF-lα expression was compared with the clinicopathologic parameters related to STAT3. Results The expression level of HIF-let and STAT3 was higher in GC tissue than in normal tissues (P 〈0. 05 ,respectively). The expression of HIF-lct and STAT3 increased with lymph node metastasis and TNM stage (P 〈 0. 05, espec- tively). The expression of HIF-lct and STAT3 was not associated with age, sex or with location of lesions (P 〉 0. 05, respectively). Conclusion The expression level of HIF-lα and STAT3 is closely related to the biologi- cal behavior of GC.
