摘要
目的 观察实验性草酸钙结石形成时肾组织中维生素K(VK)依赖性羧化酶活性变化并探讨其意义。 方法 通过 1%乙二醇诱导大鼠草酸钙结石形成 ,提取鼠肾微粒体蛋白 ,在〔14 C〕NaHCO3 参与下进行体外羧基化反应 ,检测VK依赖性羧化酶活性。 结果 形成草酸钙结石鼠肾组织中VK依赖性羧化酶活性明显增强 ,羧基化反应时加入草酸钙或外源性底物亦可使此酶活性增强。 结论 草酸钙能增强VK依赖性羧化酶的活性 。
Objective To study the activity of vitamin K dependent carboxylase in an experimental stone forming condition. Methods Rat calcium oxalate urolithiasis was induced by feeding 1% ethylene glycol in drinking water for 20 days.The microsomal protein was prepared from kidney tissue.The activity of carboxylase was detected after a carboxylation reaction in vitro. Results The activity of vitamin K dependent carboxylase was higher in rats with experimental urolithiasis.It was also stimulated by calcium oxalate in vitro. Conclusions Vitamin K dependent carboxylase activity is enhanced by calcium oxalate lithiasis.It suggests that the carboxylase may play a role in the pathogenesis of urolithiasis.
