保定地区肾综合征出血热病人血清PCR分型及核苷酸序列测定

Detection of Serun Type by PCR and Necleotile Sequcnce of Viralstrains in Patients with Hemouuhagic Fever with Renal Syndrone in Baoding

为明确保定肾综合征出血热 (HFRS)疫区的型别 ,采用RT PCR方法 ,检测急性期HFRS病人血清的汉坦病毒(HV)RNA ,并进行分型及部分核苷酸序列的测定 ,并与国内外HV的代表株进行比较。结果 31份急性期病人血清经RT PCR分型 ,2 4份为汉城型 (SEO型 ) ,2份为汉滩型 (HTN型 ) ,并测得SEO型C3株M基因 (位于 1985— 2 314位 ) ,HTN型LBY株M基因 (位于 1996— 2 314位 )核苷酸序列与国内外代表株的核苷酸和氨基酸序列相比较 :C3与 76 118、A9、LBY核苷酸同源性分别为 :6 8 79%、6 7 5 8%、6 8 0 3% ;氨基酸同源性分别为 :77 2 7%、77 2 7%、78 3%。C3与Seoul、R2 2相比 ,核苷酸同源性分别为 :95 15 %、93 94% ;氨基酸均为 97 2 7%。LBY与 76 118、A9、Seoul、R2 2核苷酸同源性分别为 :98 75 %、87 15 %、6 8 34 %、6 8 0 3% ;氨基酸同源性分别为 :10 0 %、96 2 3%、78 3%、78 3%。首次发现与韩国 76 118株同型(HTN型 )HV在保定的存在 。

To study Hantaan viral types in baoding Methods:RT Nested PCR was used to determinate Hantann viral (RNA and serum type)in serum samples from 31 acute patients with Hermorrhagic fever with renal syndrome.Some viral nedeic avid sequences were detected and compared with Hantaan viral standard strains reported athome and abrod.IN 31 acute patients with HFRS 24 serum samples were positire for seoul type:2 were poositive for Hantaan type.M gene of seoul type C3 strain was in 1985 2314 region.Compared with 76 118,A9 and LBY.the honologies of nucleic avid sequence of C3 strain were 68 79%,67 58%,68 03% respectively,amino avid sequence were 77 27%,77 27%,78 3% respectively.Compard with seoul and R22,the honologies OF nucleic avid sequence were 95 15%,93 94% respectively,amino avid aequences were 97 27%,97 27%.Compard with 76 118,A9.Seoul and R22,the homologies of LBY strain were 98 75%,87 15%,68 34%,68 03% respectively,amino and sequence were 100%,96 23%,78 3%,78 3%.Conclusion:It was certain firstly that was 76 118 subtype of Hantaan tye in Baoding and Hantaan type in Baoding
