内毒素诱导实验性暴发性肝功能衰竭模型中肝细胞再生程度分析 被引量:1

摘要 目的 :检测再生肝细胞表面增殖细胞核抗原 (PCNA)表达分布状态 ,对肝细胞再生程度作出动态评价。方法 :制备亚急性暴发性肝功能衰竭模型 ,以 PCNA单克隆抗体 ,采用免疫组织化学技术检测。结果 :大白鼠术后不同时期 ,AL T、TB水平呈进行性增高 ,术后 2 4h PCNA阳性细胞百分率进行性下降 ,与对照组相比分别为 2 4h(18.8± 11.2 ) % / (30 .7± 16 .8) % (P <0 .0 5 ) ,36 h (7.7± 5 .7) % / (2 8.5± 12 .3) % (P <0 .0 1) ,48h(5 .7±8.9) % / (2 5 .8± 8.9) % (P <0 .0 1) ,72 h(2 .8± 6 .4) % / (15 .6± 7.2 ) % (P <0 .0 1)。结论 :实验性亚急性暴发性肝功能衰竭模型中肝细胞的再生进行性受到抑制 ,可能与机体免疫因素参与有关。 Objective:Our aims were to determine the expression of proliferating cellular nuclear antigen(PCNA) in surface of the regenerative hepatocyte and to evaluate the development of the hepatocyte regeneration. Methods:We established experiment model of subacute fulminant hepatic failure(FHF) and used immunohistochemical staining technique. Results:We found ALT and TB level increased gradually in the different stage after operation in the FS 112 treated compared with the controls. The labeling index of PCNA decreased gradually at 24 hours after the operation in FHF rats compared with the controls. They were(18.8±11.2)%/(30.7±16.8)% at 24 hour(P<0.05), (7.7±5.7)%/(28.5±12.3)% at 36 hour(P<0.01),(5.7±8.9)%/(25.8±8.9)% at 48 hour(P<0.01),(2.8±6.4)%/(15.6±7.2)% at 72 hour(P<0.01). Conclusion:The hepatocyte regeneration was inhibited. It may be related to the immunologic factors of the body.
