HPLC法测定养血安神糖浆中2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷和毛蕊花糖苷的含量

Determination of 2,3,5,4'-Tetera-hydroxystilbene-2-O-β-D-glucoside and Verbascoside in Yangxue Anshen Syrups by HPLC

目的:测定养血安神糖浆中毛蕊花糖苷和2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量。方法:采用HPLC法同时测定毛蕊花糖苷和2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量。色谱柱:SHISEIDO CAPCELL PAK C_(18)柱(250mm×4.6 mm,5μm);流动相:乙腈-甲醇-1%乙酸溶液(10:13:77);检测波长:2,3,5,4'-四羟基二苯乙烯2-O-β-D葡萄糖苷:320 nm、毛蕊花糖苷:334 nm;柱温:35℃;流速:0.8 ml·min^(-1);进样量:10μl结果:2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷:线性范围为0.40μg·ml^(-1)～0.16 mg·ml^(-1),r=0.999 9,平均回收率为100.03%,RSD为1.32%(n=6);毛蕊花糖苷:线性范围为0.40μg·ml^(-1)～0.16 mg·ml^(-1),r=0.999 9,平均回收率为103.96%,RRD为0.82%(n=6)。结论:方法简便、快速、准确,为提高和完善该制剂的质量标准提供了依据。

Objective：To determine the content of 2,3,5,4＇-tetera-hydroxystilbene-2-O-β-Z）-glucoside and verbascoside in Yangxue Anshen syrups.Method：2,3,5,4-Tetera-hydroxystilbene-2-O-β-Z）-glucoside and verbascoside were determined on a SHISEIDO CAPCELL PAK C_（18）（250 mm×4.60 mm,5μm） column using acetonitrile-methanol-1%acetic acid solution（10：13：77） as the mobile phase,334nm and 320nm as the detection wavelength,35℃as the column temperature and 0.8 ml·min^（-1） as the flow rate.Result： The linear range of 2,3,5,4＇-tetera-hydroxystilbene-2-O-β-Z）-glucoside was 0.40μg·ml^（-1）-0.16 mg ？ ml^（-1）（r =0.999 9）. The average recovery was 100.03%（RSD = 1.32%,n =6）.The linear range of verbascoside was 0.40μg·ml^（-1）-0.16 mg ？ ml^（-1）（r = 0.999 9）.The average recovery was 103.96%（RSD =0.82%,n =6）.Conclusion：The method is simple,accurate and repeatable, and can provide the basis for the improvement and optimization of the quality standard of Yangxue Anshen syrups.
