摘要
目的探讨姑息期血瘀证大肠癌血清差异表达的蛋白质及意义。方法选取姑息期血瘀证、气血亏虚证大肠癌患者各6例(血瘀证组、亏虚证组),另择同期健康平衡体质者6例(对照组),通过双向凝胶电泳以及质谱鉴定和生物信息学分析,寻找血瘀证组患者血清中特异表达的蛋白质。结果血瘀证组出现7种差异表达的蛋白质:在血瘀证及气血亏虚证中均升高的蛋白质为触珠蛋白、IL-8、转移相关蛋白、血管生长因子受体-1,均降低的为血清淀粉样蛋白A;在血瘀证中高表达而在气血亏虚证中低表达的是载脂蛋白A—1前体;而在血瘀证中低表达且在气血亏虚证中高表达的是纤维结合蛋白。结论7种差异表达蛋白质的组合性变化可能是姑息期血瘀证大肠癌的物质基础,具有潜在的诊断、评估预后或指导治疗的价值。
Objective To investigate differentially expressed serum proteins in palliative stage colorectal cancer patients with blood stasis syndrome and blood/qi deficiency syndrome. Methods Two-dimensional difference gel electrophoresis and mass spectrometry were used to screen the differentially expressed serum proteins among 18 patients with blood stasis syn- drome and blood and qi deficiency syndrome; 6 healthy subjects with yin-yang harmony served as controls. Results Inter- leukin-8 and haptoglobin were highly expressed in patients with blood and qi deficiency syndrome and blood stasis syndrome; while serum amyloid A protein precursor (SAA) was decreased in both groups. Metastasis-associated protein, apolipoprotein A-I precursor, vascular endothelial growth factor receptor-1 were highly expressed in blood stasis syndrome group while decreased in patients with blood and qi deficiency syndrome. Fibronectin was decreased in patients with blood stasis syndrome, while highly expressed in patients with blood and qi deficiency syndrome. Conclusion The proteins identified in this study may be related to the development of blood stasis syndrome in patients with colorectal cancer and might be used as molecular targets for diagnosis and treatment of the syndrome.
