摘要
以脂质体转染技术构建了表达鸡马立克氏病毒(MDV)gB基因的重组痘苗病毒(RVV-gB),表明该病毒在CV-1(猴肾细胞)细胞系内能稳定传代,经腹腔 1×10(~6) PFU·羽^(-1)将重组病毒、HVT冻干苗及痘苗病毒分别按试验程序,免疫1日龄SPF鸡,并于15日龄以3×10~2PFU·羽^(-1)MDV强毒株GA株攻毒,重组病毒和HVT冻干苗匀获得较高的保护,结果证明了gB是MDV的宿主保护性抗原.此研究为CTL应答检测奠定了基础.
The recombinant vaccina virus (RVV) expressing Glycoproteins B of Marek's disease virus (MDVgB) were constructed by Lipofectin Regeant transfection This virus can be passaged stably in CV-1 cells. One-day-old SPF chickens were inoculated intraperitoneally with dose, 20~6 PFU of RVV, HVT freeze-drying-vaccine and vaccina virus respectively according to the procedure of experiment, and were challenged with dose 3 × 10~2 PFU of virulent MDV strain GA in their fifteen-days. High protection were acquired of both RVV and HVT freeze-drying-vaccine.These re- sults suggest that gB is a protective antigen of host animal. This study laid a foundation for cytotoxic T cell (CTL) dec- tection.
