糖尿病大鼠肝纤维化对血糖变化的影响及其机制 被引量:1

摘要 目的研究糖尿病所致大鼠肝纤维化对血糖变化的影响及其机制。方法将大鼠随机分为正常组1、2、3,每组12只;模型组1、2、3,每组12只。除正常组外,其余各组用STZ(链脲佐菌素)诱导并建立糖尿病SD大鼠模型,将造模成功的DM大鼠随机抽样分成以下3个组:模型组1、2、3,对组1、2、3分别干预处理8周、12周、16周,放射免疫测定肝纤维化指标:透明质酸、IV型胶原,测定空腹、餐后2h血糖;免疫组织化学法检测肝组织TGF-β1表达。结果模型组肝纤维化指标明显增高,以模型组3为主;模型组空腹和餐后血糖波动明显,肝脏TGF-β1表达显著增高,与正常组比较差异有统计学意义(P<0.05)。并且以模型组3差异最显著(P<0.01)。结论糖尿病大鼠肝纤维化对血糖有显著影响,且随病变加重而明显,其影响可能与TGF-β1在肝组织的表达增多有关。 Objective To study the effects of the change of the blood glucose on the diabetes-induced liver fibrosis rats. Methods The rats were randomly divided into 6 groups: normal control group 1, 2, 3, and the model group 1, 2, 3. Each group of rats were intervened for 8, 12 and 16 weeks by the necessary measures. Hyaluronan and Type IV collagen of the indexes of liver fibrosis were detection by radiative-immune technology. Fasting plasma glucose and 2-hour postprandial plasma glucose were measured. The expression of TGF-β1 in the liver were observed with immunohistochemical assay. Results HA.IV-C in the model group were significantly thickened than these in the normal groups (P 〈 0.05), especially in the model group 3. Model group lead to wide fluctuations in FPG and PPG. And the expression of TGF- [3 ~ in the model groups were statistically significantly (P 〈 0.05) than that in normal control group, especially in the model group 3(P 〈 0.01). Conclusion Significant effect of the liver fibrosis of diabetic rats were on FPG and PPG. The wide fluctuations of the plasma glucose may be associated with upregulation of TGF-β1 in the hepatic tissue of model groups.
