透明质酸钠对兔骨关节炎软骨VEGF及其受体-2mRNA表达的影响 被引量:3

Effect of Sodium Hyaluronate on Vascular Endothelial Growth Factor and Its Receptor-2 mRNA Expression in Rabbit Osteoarthritic Cartilage

摘要 目的:观察透明质酸钠(HA)关节腔注射对创伤性骨关节炎(OA)模型关节软骨VEGF及受体-2(VEGFR-2)mRNA表达的影响。方法:24只大耳白兔随机分为A、B、C 3组,A组为正常对照组,B、C两组行单膝前交叉韧带切断术,4周后B组关节腔内注射0.3ml生理盐水药物;C组关节腔注射等量的高分子量1%透明质酸钠(HA),每周1次,连续5周。术后11周处死动物,比较各组股骨内髁关节软骨的大体变化,采用逆转录-聚合酶链反应(RT-PCR)和实时定量聚合酶链反应(Real-Time PCR)方法检测软骨VEGF及受体-2mRNA的表达水平。结果:大体评分显示生理盐水注射组软骨退变明显重于HA注射组;VEGF及VEGFR-2mRNA的表达量较正常对照组明显升高(P<0.05),B、C两组VEGF mRNA的表达没有差别,C组VEGFR-2mRNA水平较B组显著下降(P<0.05)。结论:VEGF及VEGFR-2mRNA表达在OA软骨中表达明显升高,关节腔注射HA对软骨VEGFmRNA表达没有抑制作用,但可以通过抑制VEGFR-2mRNA表达来减轻软骨退变的程度。 Objective: To observe the influence of intra-articular injection of sodium hyaluronate (HA) on mRNA expression of vascular endothelial growth factor (VEGF) and its receptor-2 (VEGFR- 2) in cartilage of rabbit osteoarthritis (OA) model. Methods: Twenty four white rabbits were di- vided into A,B,C groups randomly. Group A were normal control group, B and C groups were underwent unilateral anterior cruciate ligament transection (ACLT). The rabbits in group B were injected normal saline after ACLT, Group C received intra-articular 1% sodium hyaluronate (HA) injection 5 weeks after surgery, for 0.3 ml once a week. In the 6th week after surgery, all the rabbits were sacrificed. The cartilage changes on the medial femoral condyles were graded separately, mRNA expression of VEGF and VEGFR-2 were detected by using reverse transcrip- tion-polymerase chain reaction (RT-PCR) and real time polymerase chain reaction (Real Time-PCR). Results. Cartilage degeneration in untreated group was significantly severer than that in HA injection group. Intraarticular 1% sodium hyaluronate injection inhibited VEGFR-2 expres- sion but had no effect on reducing the VEGF mRNA expression in cartilage. Conclusion. These results suggested that VEGF and VEGFR-2 may be involved in the progression of OA and in the therapeutic mechanism of HA at least partly through the influence of VEGFR-2 expression during the development of OA.
