白藜芦醇对鼻咽癌细胞ATP酶抑制作用研究

摘要 目的检测30μmol/L白藜芦醇对鼻咽癌细胞ATP酶活力及蛋白表达抑制作用。方法超微量ATP酶试剂盒检测白藜芦醇对鼻咽癌细胞膜的Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶和T-ATP酶活力的变化;Western-blot检测白藜芦醇对鼻咽癌细胞ATP酶蛋白表达差异。结果 RES能显著抑制多种鼻咽癌细胞Na+-K+-ATP酶活力(P<0.01)、Ca2+-Mg2+-ATP酶活力(P<0.01)和T-ATP酶活力(P<0.001),Western-blot检测结果显示白藜芦醇能浓度依赖性下调ATP的表达(P<0.05)。结论白藜芦醇抑制鼻咽癌细胞株ATP酶活力,显著下调ATP酶蛋白表达,可能是抑制鼻咽癌细胞株增殖的分子机制之一。 Objective To detect the activities inhibition of ATPase and protein expression by 30 μmol/L Resveratrol in nasopharyngeal carcinoma cells. Methods The activities of NPC cells' ATPase were measured by ultramicro ATPase Kit, the proteins expression of ATP was determined by western blot. Results RES could significantly inhibit activities of Na+-K+-ATPase(P 〈 0.01), Ca2+-Mg+-ATPase(P 〈 0.01) and T-ATPase(P 〈 0.001) in NPC cells. Western blot suggested RES down-regulated the expression of ATP (P 〈 0.05). Conclusion The molecular mechanisms for RES to inhibit the proliferation of nasopharyngeal carcinoma cell lines could be inhibition of activities of ATPase and down- regulation of the expression levels of ATPase protein.
