化浊解毒汤对实验性大鼠胃癌前病变的实验研究 被引量:3

摘要 目的:观察化浊解毒汤对实验性大鼠胃癌前病变癌基因表达的影响,探讨化浊解毒汤对胃癌前病变治疗作用机理。方法:将50只雄性大鼠随机分为空白组、模型组、胃复春组、中药高剂量组、中药低剂量组,每组10只。除空白组其余4组造模成功后开始治疗,中药治疗组给化浊解毒汤,西药治疗组给予胃复春,共4周。末次给药后禁食、水24 h,称重,麻醉后处死,测各组大鼠胃黏膜癌基因的阳性表达率。结果:各治疗组与模型组,经治疗后癌基因Bcl-2、c-myc的阳性表达率均降低,差异有显著性(P<0.05)。中药高剂量组与模型组对比,癌基因Bcl-2、c-myc的阳性表达率明显降低,中药低剂量组与西药组相比均差异无显著性(P>0.05)。结论:化浊解毒汤通过降低癌基因Bcl-2、c-myc蛋白的阳性表达,从而恢复细胞增殖与凋亡之间的平衡状态,是该方治疗胃癌前病变机制之一。 Objective: to observe the effect of Huazhuo Jiedu decoction on oncogene expression in precancerous lesion of stomach,and to explore its functional mechanism in treating lesion before gastric cancer.Methods: 50 male rats were randomly divided into control group,model group,Weifuchun group,high dose group of Chinese medicine and low dose group(n=10).Except control group,all others were treated after models made,Chinese medicine groups administered with Huazhuo Jiedu decoction while western medicine group with Weifuchun,for 4 weeks.The last administration was followed by fasting and water depriving for 24 hours.Weighing and killing the rats with anesthesia to detect the positive expression rate of gastric mucous membrane oncogene.Results: Bcl-2 and c-myc positive expression rate of the treatment groups and model group all decreased with significant difference(P<0.05).Compared to model group,the positive expression rate of the high dose group significantly decreased while there was no significant difference between low dose group and western medicine group(P>0.05).Conclusion: Huazhuo Jiedu decoction is able to restore a balance between cell multiplication and apoptosis by decreasing the positive expression of Bcl-2 and c-myc,which is one of the mechanisms in treating precancerous lesion of stomach.
