摘要
目的研究TRAIL和紫杉醇联合应用对骨肉瘤细胞的抑制作用,寻找骨肉瘤临床治疗的新方案。方法将TRAIL与紫杉醇单独或联合应用于体外培养的骨肉瘤OS-732细胞,采用MTT法测定其对OS-732细胞的抑制率,并于相差显微镜、荧光显微镜下观察细胞形态的改变。结果 50 ng/mL的TRAIL与10μg/mL紫杉醇联合作用于OS-732细胞24 h后,抑制率为47.49%,明显高于单用TRAIL(50 ng/mL)时的9.85%及单用紫杉醇(10μg/mL)时的20.37%(P<0.01)。细胞形态结构均出现凋亡的特征性变化。结论 TRAIL与紫杉醇联合应用可高效杀伤骨肉瘤细胞,这种杀伤效应是通过促进OS732细胞的凋亡来实现的。
[Objective] To study the inhibition effect of TRAIL combined with Paclitaxel on the osteosar-coma cell lines so as to explore the new method of the clinical treatment. [Methods] TRAIL and Paclitaxel were used on the OS-732 cells individually or jointly so as to observe their synergistic effects on the apopto-sis of osteosareoma cells. [Results] Twenty-four hours after the application of 50 ng/mL TRAIl, and 10 μg/ml, Paclitaxel jointly on OS-732 cells, The inhibition rate is 47.49% by MTT essay which is significantly higher than the application of 50 ng/mL TRAIL(9.85%) and 10 μg/mL Paclitaxel (20.37%) respectively (P〈0.01). Mor-phological changes observed with inverted phase contrast microscope, fluorescence microscope showed tumor-killing effect of joint application was much stronger than respective application. [Conclusion] Combination of TRAIL and etoposide is obviously conducive to killing effect of osteosarcoma cells which were achieved by inducing apoptosis.
