双歧杆菌脂磷壁酸对刀豆蛋白A诱导小鼠免疫性肝损伤的保护作用

Protective effect of lipoteichoic acid extracted from bacillus bifidus against autoimmune hepatitis induced by Con A in mice

目的研究双歧杆菌脂磷壁酸(LTA)对Con A诱导的小鼠免疫性肝损伤的保护作用,探讨其在该疾病预防和治疗中的应用可行性。方法小鼠随机分为正常组、模型组、联苯双酯组(150 mg/kg)和LTA处理组(100、200和400μg/kg),尾静脉注射Con A复制小鼠免疫性肝损伤模型;观察各组小鼠的一般情况及肝、脾指数,生化法和酶联免疫法测定血清ALT、AST和TNF-α的含量;RT-PCR检测小鼠肝组织内NF-κB的基因表达。结果 LTA处理组小鼠的一般情况和肝脏指数均好于模型组,其血清内ALT、AST和TNF-α的水平显著低于模型组(P<0.05),但与联苯双酯组基本相当(P>0.05);不同浓度LAT处理组小鼠肝细胞NF-κB的mRNA水平均低于模型组(P<0.05),且LTA 400μg/kg组与联苯双酯组基本相当(P>0.05)。结论双歧杆菌脂磷壁酸的双向免疫调节作用可以有效保护Con A诱导的小鼠免疫性肝损伤。

[Objective] To explore the protective efiect of lipoteic, hoic acid （LTA） extracted from bacillus bifidus on the autoimmune hepatitis of mice model injected by Con A. [Methods] The mice were divided in-to six groups including control group, Con A model group, bitendate group and groups treated with different concentrations of LTA （100 μg/kg, 200 μg/kg, 400 μg/kg）. The models of autoimmune hepatitis were indueed by Con A injected via tail vein. The general conditions, liver and spleen index were evaluated as well as the levels of ALT, AST and TNF-a in serum. The expression of gene NF-KB of liver were also assayed by the means of RT-PCR. [Results] Compared with ttle Con A model group, the general conditions and the liver index of groups treated with LTA were better, and the levels of AST, ALT, TNF-α in serum and the mRNA of NF-KB of liver were significantly lower （P 〈0.05）; however, the indexes above of LTA 400 μg/kg group were similar to bifendate group. [ Conclusion] The LTA extracted from bacillus bifidus ean effectively proteet the mice of autoimmune hepatitis induced by Con A, and it is worthy to be studied further in the precaution and therapy to the patients with this disease.
